摘 要: | 目的黄连吴茱萸配伍传统丸剂与现代临床水煎剂治疗高脂血症模型大鼠,研究两种剂型对高脂血症大鼠降血脂作用的差异性。方法高脂乳剂灌胃建立大鼠高脂血症模型,丸剂、水煎剂灌胃给药,采用酶法测定血清TC、TG、LDL、HDL含量,采用实时荧光定量PCR技术检测肝脏HMG-Co A还原酶、肝脏型脂肪酸结合蛋白基因的表达。结果模型对照组血清中TC、TG和LDL含量显著高于空白对照组,丸剂、水煎剂组上述指标均较模型对照组显著降低,有显著的统计学意义(P0.05);模型对照组肝脏HMG-Co A还原酶、肝脏型脂肪酸结合蛋白基因较空白对照组分别增高20.7倍、6.33倍,丸剂组较模型对照组上述基因表达分别降低3.28倍、39.08倍,水煎剂基因表达分别降低2.92倍、11.35倍。结论黄连吴茱萸丸剂、水煎剂均能降低实验性高脂血症大鼠血脂水平,抑制HMG-Co A还原酶、肝脏型脂肪酸结合蛋白基因高表达,丸剂降血脂作用优于水煎剂,基因抑制作用更显著(P0.01),提示黄连吴茱萸配伍传统丸剂在降血脂方面更有优势且更具科学性。

Study on the Effect of Different Dosage Forms of Coptis-Evodia Herb Couple on Reducing Blood Lipid |

Abstract: | Objective To study the effect of two kinds of dosage forms on reducing blood lipid in hyperlipemia rats, the traditional pills and modern clinical decoctions were used for the treatment of hyperlipidemia model rats, respectively. Methods Hyperlipidemia rats induced by fat emulsion, meanwhile TC, TG, LDL and HDL in serum were measured by the enzymatic method. Additionally the expression of HMG-CoA, L-FABP gene in liver tissues was detected by real-time PCR. Results The levels of TC, TG and LDL in serum of the model group were significantly higher than those of the control group. Compared with the model control group, pills and decoctions could reduce the levels of indicators above all, which revealed a statistical significance ( P<0. 05 ) . Additionally the expression of HMG-CoA and L-FABP gene in livers of model control group were 20. 7 and 6. 33 times, respectively, as higher as that of the control group, which were notably decreased by 3. 28 and 39. 08 times after administration of pills compared with model control group, while after administration of decoctions these genes decreased by 3. 28 and 39. 08 times. Conclusion Coptis-Evodia herb couple in pills and decoctions can reduce blood lipid levels in hyperlipidemic rats and inhibit HMG-CoA and L-FABP gene expression. What’ s more, Coptis-Evodia herb couple in pills was much better than decoctions in gene inhibition. The survey indicated that Coptis-Evodia herb couple in pills was more advantageous and scientific. |
