99Tc-MDP对大鼠佐剂性关节炎PGE1和PGE2的影响

目的    观察⁹⁹Tc-MDP对佐剂性关节炎（AA）大鼠模型的影响，探讨⁹⁹Tc-MDP在类风湿关节炎（RA）治疗中的抗炎作用及其机制。方法    采用完全性弗氏佐剂足跖皮内注射法建立AA模型，随机将30只雄性Wistar大鼠分为正常对照组、AA模型组和⁹⁹Tc-MDP治疗组，治疗组在造模10d后给予⁹⁹Tc-MDP2.5mL/kg尾静脉注射治疗，1次/d，治疗15d；正常对照组和AA模型组给予相同体积的生理盐水尾静脉注射治疗，1次/d，治疗15d。观察大鼠左踝关节宽度、关节炎指数（AI），以及应用酶联免疫吸附法（ELISA）检测血清中的PGE1和PGE2的水平。结果    ①AA模型组、⁹⁹Tc-MDP治疗组随着免疫时间的延长，左踝关节肿胀程度逐渐减轻， ⁹⁹Tc-MDP治疗组较AA模型组减轻明显（P＜0.05）；AA模型组和⁹⁹Tc-MDP治疗组关节炎指数逐渐下降，⁹⁹Tc-MDP治疗组较AA模型组下降明显（P＜0.05）；②各组血清中PGE1水平差异无统计学意义（P＞0.05）; AA模型组、⁹⁹Tc-MDP治疗组血清中PGE2水平均高于正常对照组（P＜0.05），AA模型组高于⁹⁹Tc-MDP治疗组（P＜0.05）。结论    ⁹⁹Tc-MDP可以降低AA大鼠左下肢踝关节宽度、关节炎指数及血清中的PGE2的水平，对抑制AA大鼠的炎症有一定疗效。

Effects of 99Tc-MDP on PGE1 and PGE2 in rats of adjuvant arthritis

Objective     To observe the effects of ⁹⁹Tc-MDP (technetium-99 conjugated with methylene diphosphonate) on adjuvant arthritis in rats, and study its anti-inflammatory mechanism. Method    The AA(adjuvant arthritis) rat model was established by intradermal injection of complete Freud′s adjuvant. Thirty male Wistar rats were divided randomly into three groups: control group, adjuvant arthritis group and ⁹⁹Tc-MDP treatment group. In treatment group,  ⁹⁹Tc-MDP （2.5mL/kg）was administered  through caudal vein on the 10th day and repeated 15 days later after arthritis was induced. Same volume normal saline was given to the other two groups under the same protocol. Then, left-right diameter of left hind ankle, arthritis index of each rat were recorded while  PGE1 and PGE2 in peripheral blood were measured by ELISA. Results    ①The left right diameter of left hind ankle in both  control group and  ⁹⁹Tc-MDP treatment group reduced with time, but the diameter in  ⁹⁹Tc-MDP treatment group significantly reduced（P＜0.05）than that of AA group； The arthritis index of AA group and ⁹⁹Tc-MDP treatment group both decreased with time, but the index  decreased much more significantly（P＜0.05）in the latter. ②There were no significant differences among all groups on the level of PGE1（P＞0.05）,and the level of PGE2 of the AA group and  ⁹⁹Tc-MDP treatment group were notably higher than the control（P＜0.05）. The PGE2 level of control l group was much higher than that of ⁹⁹Tc-MDP treatment group（P＜0.05）. Conclusion      ⁹⁹Tc-MDP  can reduce the left-right diameter of hind ankle and arthritis index in adjuvant arthritis rats.  ⁹⁹Tc-MDP may have a certain anti-inflammatory effect on adjuvant arthritis rats.
