摘    要: 目的探讨人甲状旁腺激素1-34[human parathyroid hormone(1-34),hPTH1-34]对去卵巢大鼠骨质疏松症的治疗作用。方法 分别予去卵巢6周的Wister大鼠hPTH1-34(40μg·kg-1·d-1,sc每周5次)或生理盐水干预8周,假手术大鼠不予治疗。留取 腰椎1-6(L1-6)、右侧股骨及血、尿标本分别进行骨密度、骨生物力学和生化指标的测定。结果 hPTH1-34组L1-6、股骨远端和股 骨干骨密度值明显高于去卵巢对照组,且L1-6和股骨干的骨密度值与假手术组无差异;凹入试验中hPTH1-34组Can.Load和 Can.Stiff明显高于去卵巢对照组,和假手术组无差异;三点弯曲实验中hPTH1-34组E.Modulus值与对照组无统计学差异;hPTH1-34 组血ALP和UDpd/Cr水平低于去卵巢对照组,但高于假手术组。结论 hPTH1-34可以明显增加去卵巢骨质疏松大鼠的骨量和 骨生物力学特性。

Abstract:OBJECTIVE To investigate the effect of human parathyroid hormone fragment-(1-34) treatment on bone mass and biomechan-ical properties in ovariectomized rats.METHODS The ovariectomized rats were treated for 8 weeks with hPTH1-34 group (40μg·kg-1·d-1, sc, five times per week) or vehicle as OVX control group respectively, the sham-ovariectomized rats were untreated. All of the samples of lum-bar vertebrae(L1-6), right femur, blood and urine were collected while rats were sacrified. The BMD of lumbar vertebrae and femur ,the parameters of femoral biomechanics and the levels of bone turnover markers were measured. RESULTS The BMD values at L1-6, distal femur and femoral shaft in hPTH1-34 group were significantly increased respectively compared with those in OVX control group. No significant differences were observed in BMD values of lumbar, femoral shaft between hPTH1-34 group and sham group. The values of Can. Load and Can. Stiff at distal femur in hPTH1-34 group were higher than those in OVX control group respectively. No significant differences were observed in the values of Can.Load and Can.Stiff at distal femur between hPTH1-34 group and sham group. The values of Max.Load at femoral shaft in hPTH1-34 group were higher than those in sham group and OVX control group. There were no significant differences in the values of elastic modulus at femoral shaft among hPTH1-34 group, OVX control group and sham group. The levels of serum ALP and urine Dpd/Cr in hPTH1-34 group were lower than those in OVX control group, while higher than those in sham group. CONCLUSION hPTH1-34 has a pronounced anabolic effect on bone mass and biomechanical properties in ovariectomized osteoporotic rat model.
