Omi/HtrA2在食管癌中的表达及其与细胞凋亡的关系

目的探讨Omi/HtrA2蛋白在食管鳞癌中的表达及其与肿瘤细胞凋亡的关系。方法采用免疫组化SP法对95例经病理确诊为食管鳞癌患者的正常组织、癌旁组织及癌组织进行Omi/HtrA2蛋白半定量免疫组化分析,用TUNEL法检测石蜡切片中肿瘤细胞凋亡情况,通过统计学软件SPSS 11.0进行结果分析,确定食管癌中Omi/HtrA2表达对肿瘤细胞凋亡的影响。结果①Omi/HtrA2在食管正常组织、癌旁组织、鳞癌组织的阳性表达率分别为22.50%、27.50%、74.74%。②Omi/HtrA2蛋白在不同组织中的表达情况进行两两比较,发现癌组织的阳性率与正常组织、癌旁组织比较,有显著性差异（P〈0.05）,正常组织与癌旁组织阳性率比较未见有显著性差异,无统计学意义（P〉0.05）。③Omi/HtrA2表达阳性者细胞凋亡指数明显高于Omi/HtrA2表达阴性者（P〈0.01）。结论 Omi/HtrA2参与了食管鳞癌的发生、发展,可以作为判断食管鳞癌预后的指标,Omi/HtrA2在食管癌细胞的凋亡中发挥凋亡促进作用。

The expression of Omi/HtrA2 and its relationship with apoptosis in esophageal cancer

To investigate the expression of Omi/HtrA2 protein in esophageal squamous carcinoma and its relationship with the apoptosis tumor cell. Methods With the immunohistochemical SP method, carried through the semiquantitative immunohistochemical analysis of Omi/HtrA2 protein to the normal tissue, adjacent tissue and cancer tissue in 95 patients who were pathologically confirmed suffering the disease of esophageal squamous carcinoma. Using the TUNEL to check the apoptosis of tumor cells in paraffin. Analyzed the results with the statistical software SPSS 11.0, determined the effects of the expression of Omi/HtrA2 to the tumor cell apoptosis in esophageal carcinoma. Results ①The positive expression rate of Omi/ HtrA2 in normal esophageal tissues, adjacent tissues, squamous carcinoma was 22.50%, 27.50%, 74.74% respectively. ②The pairwise comparison of Omi/HtrA2 protein in different tissues, comparing the positive rate between the cancer tissue and the normal tissue, adjacent tissues there were significant difference（P〈0.05）, comparing the normal tissue and adjacent tissues, the result had no statistically significant difference（P〈0.05）. ③The apoptotic index of Omi/HtrA2 positive expression was significantly higher than the Omi/HtrA2 negative expression（P0.01）. Conclusion The Omi/HtrA2 is involved in the occurrence and development of esophageal squamous carcinoma, and can be used as an indicator to determine the prognosis of esophageal squamous cell carcinoma. The Omi/HtrA2 plays an promotive role in apoptosis of esophageal cancer.
