纳米砷与三氧化二砷的体外细胞毒性比较研究

摘    要:目的通过比较三氧化二砷与纳米元素砷两种砷制剂在体外对肺腺癌细胞株以及正常小鼠淋巴细胞的影响,对纳米砷制剂的作用进行初步评估。方法将不同浓度的纳米砷或三氧化二砷与肺腺癌细胞株A549或H1299细胞体外作用2d,将不同浓度的纳米砷或三氧化二砷与小鼠脾细胞作用24h,将不同浓度的纳米砷或三氧化二砷加白细胞介素2与小鼠脾细胞作用72h。随后以MTT检测法分别检测各组的杀伤率。结果纳米砷与三氧化二砷对肺腺癌细胞株A549或H1299的杀伤率,在相同摩尔药物浓度下很接近。三氧化二砷与纳米砷对小鼠脾脏淋巴细胞与rIL-2活化的淋巴细胞的毒性作用也相当接近,且三氧化二砷与纳米砷在低浓度1.5umol/L时,其杀伤率巳达40%以上,二者差异无显著性(P〉0.05)。结论纳米元素砷具有较强抗癌活性,但其对正常淋巴细胞的毒性与三氧化二砷相比未见减轻。这对于笔者加深对纳米元素的认识,具有一定的意义。

Comparison of toxicities between arsenic trioxide and nanometer arsenic to lung cancer cells and normal lymphocytes in vitro

Abstract:[Objective] To evaluate the effects of nanometer arsenic by means of an in vitro experiment contrasting the effects of arsenic trioxide and nanometer arsenic on lung adenocarcinoma cell lines and normal lymphocytes. [Methods] Lung adenocarcinoma cell line A549 or H1299 were cultured in various concentrations of nanometer arsenic or arsenic trioxide for two days, normal marine spleen cells were cultured in various concentrations of nanometer arsenic or arsenic trioxide for 24 hours, marine spleen cells were cultured in rIL-2 and various concentrations of nanometer arsenic or arsenic trioxide for 72 hours, the cytotoxicity of each group was determined with colorimetric MTT assay. [Results] The cytotoxicities of nanometer arsenic to lung adenocareinoma cell line A549 or H1299 were very close to those of arsenic trioxide under the same molar concentration, the cytotoxicities of nanometer arsenic to normal murine spleen cells and alL-2 activated marine spleen cell were close to those of arsenic trioxide under the same molar concentration. Furthermore, the cytotoxicities of the both drugs to the normal cells were more than 40% at the low concentration of 1.5 mmol/L, no significant difference was seen between nanometer arsenic and arsenic trioxide at the concentration. [Condusion] Nanometer arsenic had relatively strong anticancer effects, but its toxicity to normal lymphocytes was not less than arsenic trioxide. This experiment was of significance to the knowledge of nanometer elements.
