骨立对去卵巢大鼠骨质疏松症防治作用的实验研究

摘    要:运用骨立冲剂对切除卵巢大鼠复制的类似“肾虚”骨质疏松症模型进行干预,并与西药对照组结果进行比较,通过对血清中反映骨代谢的生化指标及骨小梁形态检测结果进行综合分析,证实骨立防治原发性骨质疏松症的疗效并探讨其防治机理。方法:将60只大鼠按随机原则分成假手术组(Sham组)、造模组(Model组)、骨立高剂量组(H-GSL组)、骨立中剂量组(M-GL组)、骨立低剂量组(L-GL组)和己烯雌酚阳性对照组(DES组),共6个组,每组10只。去卵巢造模后按不同要求进行干预,12周后观察各组结果并做比较分析。结果:骨立能明显提高去卵巢大鼠的雌激素水平,纠正去卵巢后骨的高转换率,以保持适度的骨转换,减少骨质丢失,并阻止大鼠骨小梁结构的破坏;骨立高剂量组与己烯雌酚组E2水平相当(P>0.05)。结论:骨立能有效阻止骨质疏松症的发生与发展,其机制可能主要是通过抑制骨高转换率而实现其作用的。

Abstract:Objective:The purpose of this investigation is to study the effects of Gu Li(GL)on osteoporosis(OP)in ovariectomized rats by doing biochemical examine of serum on bone,and compared with diethylstilestrol(DES)group,try to demonstrate the effects of Gu Li on osteoporosis and discuss its mechanism.Method:Sixty female SD rats of three month were divided into 6 groups,sham operation(Sham),ovary resection(Model),ovary resection plus High Gu Li(H-GL),ovary resection plus Middle Gu Li(M-GL),ovary resection plus Low Gu Li(L-GL),ovary resection plus Diethylstilestrol(DES).Ten rats in each group,and were fed according to various demands.Next step was to observe and compare with the results of each group after 12 weeks.Result:GL could remarkably increase content of the estradiol of ovarietomized rats,correctted the high turnover caused by ovary resection and inhibited the undermining of the trabeculea and loses of rat bone mass,and the contents of estradiol are equal between the H-GL group and the DES group(P>0.05).Conclusion:Gu Li has antiosteoporotic effects on ovariectomized rats and the mechanism was realized by inhibiting the bone tureover probably.
