胰高血糖素样肽-2联合谷氨酰胺对放射损伤后肠黏膜细胞凋亡的影响

摘    要:目的: 探讨胰高血糖素样肽-2(GLP-2)联合应用Gln对放射损伤后肠黏膜细胞凋亡的影响. 方法: 将50只小鼠随机分为正常组、放射组、GLP-2处理组(GLP-2组)、Gln处理组(Gln组)和Gln和GLP-2联合处理组(Gln/GLP-2组),每组10只.实验期为7 d.GLP-2组于实验第1~6天腹腔注射GLP-2;Gln组于实验第1~6天灌饲Gln溶液;Gln/GLP-2组给予Gln和GLP-2联合处理,方法同上.在实验第4天,各组小鼠(除正常组)接受11Gy γ-射线腹部照射.实验第7天取小肠标本,行TUNEL法检测原位细胞凋亡,并用荧光法检测肠上皮细胞Caspase-3 酶活性. 结果: GLP-2组的肠黏膜细胞凋亡指数和Caspase-3酶活性与放射组相比无显著差异,而Gln组较放射组明显降低(P<0.01);Gln/GLP-2组的细胞凋亡指数和Caspases-3 酶活性较Gln组进一步降低(P<0.01),与正常组相比无显著差异. 结论:GLP-2和Gln联合应用能明显抑制放射损伤后肠黏膜细胞凋亡.
