反相高效液相色谱法同时测定蒲公英中槲皮素和木犀草素的含量

目的：建立蒲公英中槲皮素和木犀草素含量的测定方法.方法：采用反相高效液相色谱（RP-HPLC）法,色谱柱为Agilent HC-C18柱（250 mm × 4.6 mm,5μm）;柱温：30℃;流动相：甲醇-0.4％磷酸（52∶48）,流速：1.0 ml·min-1,检测波长：360 nm.结果：槲皮素和木犀草素的浓度分别在1.68～16.80 μg·ml-1（r =0.999 7）,1.52～ 15.20 μg·ml-1 （r=0.999 8）时线性关系良好;平均加样回收率分别为98.67％和98.46％,RSD分别为2.49％和2.89％.结论：本方法测定快速,结果准确可靠,适合同时测定蒲公英中槲皮素和木犀草素的含量.

Determination of Quercetin and Luteolin in Taraxacum Herb by RP-HPLC

Objective：To establish the determination method for quercetin and luteolin in Taraxacum Herb.Methods：An Agilent HC-C18 column （250 mm× 4.6 mm,5 μm） was used at 30℃ with the mobile phase of methanol-0.4％ phosphatic acid （52∶48）.The flow rate was 1.0 ml · min-1.The detection wavelength was 360nm.Results：The linear range was within 1.68-16.80 μg · ml-1 for quercetin （r =0.999 7） and 1.52-15.20 μg · ml-1 for luteolin （r =0.999 8）,the recovery of quercetin and luteolin was 98.67％ （RSD =2.49％） and 98.46％ （RSD =2.89％）,respectively.Conclusion：The method is rapid,accurate and reliable,and suitable for the simultaneous determination of quercetin and luteolin in Taraxacum Herb.
