HIF-1α在肝癌组织中表达及临床意义

目的:探讨缺氧诱导因子-1α(H IF-1α)在肝癌组织中表达的意义。方法:采用免疫组化S-P法检测75例石蜡包埋的人体肝癌组织(术前均未行放疗及化疗)H IF-1α的表达。结果:H IF-1α主要表达于肝癌细胞浆,部分于胞核,肿瘤间质细胞也可见少量表达。肿瘤坏死区周围及肿瘤浸润边缘部的癌细胞H IF-1α表达明显增多。H IF-1α的表达强度与肝癌细胞恶性程度、淋巴结转移及预后有关,而与病人年龄、性别及原发肿瘤大小均无关。其阳性强度随癌细胞分化程度的降低、淋巴结转移、术后生存期短而增高。结论:H IF-1α可作为评估肝癌淋巴结转移和预后的重要指标之一。

Expression of Hypoxia-inducible factor-1α in Hepatocellular Carcinoma

Objective: To investigate the significance of hypoxia-inducible factor-1α(HIF-1α) expression in hepatocellular carcinoma(HCC).Methods: The expression of HBx and HIF-1α in 75 cases of formalin-fixed paraffin-embed hepatic carcinoma tissues was detected by immunohistochemistry.Results:(HIF-1α was mainly expressed in the cytoplasm and partially in the nucleus of tumor cells,but little in the cytoplasm of mesenchymal cells.HIF-1α was evidently expressed in tumor cells at the periphery of necrosis and the margin of the tumor.The expressive intensity was related to the malignancy of the cancer cell,lymph node metastasis,and prognosis,but not to the age or sex of the patients,or the size of the primary tumors.The expressive intensity increased with the decrease of the differentiation of cancer cells,lymph node metastasis and the decrease of post-operative survival time.Conclusion: The expression of HIF-1αmay serves as an important parameter in evaluating lymph node metastasis and prognosis of hepatocellular carcinoma.
