CIK细胞体外逆转多药耐药性的研究

摘    要:目的:研究CIK细胞体外逆转多药耐药(multidrug resistance,MDR)可行性。方法:用人外周血分离出的单个核细胞,在不同细胞因子诱导下培养成细胞因子诱导的杀伤细胞(cytokine-induced killer,CIK)及树突状细胞(DC),然后将其与成熟DC进行共培养,作为效应细胞,以K562敏感株与耐药细胞株为靶细胞进行体外逆转研究。流式细胞术检测靶细胞的P-gp表达及细胞内的ADR相对含量,MTT法测定其细胞毒作用,RT-PCR检测mdr-1基因耐药性逆转表达状况。结果:K562/ADR耐药细胞株ADR含量经效应细胞细胞作用后,细胞内的ADR含量分别提高了13.80%、39.50%,(P<0.01);,P-gp分别降低12.00%、42.69%,(P<0.01),mdr-1基因表达呈下调趋势,效应细胞对靶细胞的细胞毒作用分别达45.8%、78.9%,(P<0.01)。结论:CIK+K562/ADR-DC对P-gp高表达K562/ADR耐药细胞株具有特异性的细胞毒作用,有效提高了细胞内的ADR含量,下调了P-gp、mdr-1表达,使免疫效应细胞体外逆转多药耐药的效果得以显现。
