人类细胞周期蛋白D3反义表达质粒的构建

摘    要:目的:构建人类细胞周期蛋白D3(cyclin D3)反义表达质粒,为今后利用反义核酸阻断技术研究cyclin D3基因的功能提供基础。方法:运用DNA重组技术将人cyclin D3基因反向克隆到真核表达载体pcDNA3.1中,构建成cyclin D3反义表达质粒pcDNA3--ascyclinD 3。结果:通过酶切鉴定及测序分析证实,实验成功构建了人cyclin D3反义表达质粒。结论:cyclin D3反义表达质粒pcDNA3--ascyclin D3的成功构建,为进一步研究cyclin D3的生物学功能及其在肿瘤发生、发展、诊疗和预后中的作用奠定了基础。

Abstract:Objective:To construct the eukaryotic expression plasmid expressing human cyclin D3 antisense gene to study the function of cyclin D3 gene by means of antisense technology.Methods:PcDNA3-ascyclin D3 plasmid was constructed by inserting human cyclin D3 cDNA reversely to eukaryotic expression vector PCDNA3.1.Results:It was comfirmed by DNA sequence analysis that the expression plasmid of antisense cyclin D3 was successfully constructed.Conculusion:The results of this study lay the foundation for further studying the biological functions of cyclin D3 and its role in the occurrence , development, diagnosis, therapy and prognosis of tumors.
