摘    要:目的建立同时测定壮血药酒中新北美圣草苷、柚皮苷、补骨脂素和佛手柑内酯4个主要成分含量的高效液相色谱检测方法。方法采用依利特C18柱;流动相A为乙腈-甲醇(1∶2),流动相B为0.4%冰醋酸溶液,梯度洗脱;流速为1.1 mL/min;柱温为25℃;检测波长λ_1=283 nm(新北美圣草苷和柚皮苷),λ2=222 nm(补骨脂素和佛手柑内酯)。结果在优化的色谱条件下,新北美圣草苷、柚皮苷、补骨脂素和佛手柑内酯的线性范围分别为6.52~130.40μg/mL(r=0.999 7)、6.61~132.20μg/mL(r=0.999 3)、6.75~135.00μg/mL(r=0.999 6)、4.05~81.00μg/mL(r=0.999 9);4种成分的平均加样回收率(n=6)分别为96.94%、99.14%、98.09%、96.87%,RSD分别为0.75%、0.52%、1.18%、1.02%。结论该方法简便快捷,回收率、重复性良好,可用于壮血药酒中新北美圣草苷、柚皮苷、补骨脂素和佛手柑内酯的含量分析。

High-performance liquid chromatography for simultaneous determination of four components in zhuangxue yaojiu
Abstract:Objective To establish a high-performance liquid chromatography ( HPLC) method for the simul-taneous determination of four main components ( neoeriocitrin,naringin,psoralen and bergapten) in zhuangxue yaojiu. Methods The HPLC separation was performed on an Hypersil C18 column eluted,with a mobile phase consisting of acetonitrile-methanol (1∶2) as mobile phase A,and 0. 4% acetic acid glacial solution as mobile phase B. The flow rate was 1. 1 mL/min. The column temperature was at 25 ℃. Neoeriocitrin and naringin were detected at 283 nm,and psor-alen and bergapten were detected at 222 nm. Results The chromatographic peaks of 4 main components and the corre-sponding spectra were ideal under the optimal conditions. Neoeriocitrin,naringin,psoralen and bergapten had good line-arity at 6. 52~130. 40 μg/mL (r =0. 999 7),6. 61 ~132. 20 μg/mL (r =0. 999 3),6. 75 ~135. 00 μg/mL (r =0. 999 6),4. 05~81. 00 μg/mL(r=0. 999 9),and the average recoveries and RSD were 96. 94% (0. 75%),99. 14%(0. 52%),98. 09% (1. 18%) and 96. 87% (1. 02%),respectively. Conclusion The method is simple,economical and accurate with good reproducibility for the determination of neoeriocitrin,naringin,psoralen and bergapten.
