Experimental Study on Protective Effect of Ischemic Preconditioning for Donator Liver

Objective To establish a stable transplantation model,analyze the protection effect of ischemic preconditioning on miniature banna pig＇s donor liver.Methods Original liver transplantation experiment model of miniature pigs was established.Twenty banna pigs（male or female,of the same maternal strain） were randomly divided into three groups,i.e.normal control group,ischemia reperfusion group and ischemic preconditioning group.Expression of heat shock protein 70（HSP 70） was detected by Western blotting method.The activity change of serum aspartate transaminate（AST） and alanine transaminase（ALT） were tested by the rate method on the Biochemical Analyzer.Specimen sections were prepared to observe the morphological changes of cells under the light microscope.Results Among the three groups,HSP 70 expression in ischemic preconditioning group was significantly increased,and the structure of hepatic cells had little changes.Compared with the ischemic preconditioning group,the indexes of ischemia reperfusion group had opposite changes with higher level of serum transaminase（P0.01）,lower HSP 70 expression and obvious changes in the structure of hepatic cells.Conclusion In the liver transplantation surgery on miniature banna pigs,ischemic preconditioning has protective effect on liver and positive modulation on HSP 70 expression.
