摘    要:将含灭活FBL-3瘤细胞的聚氨酯海绵块植入免疫的C57BL/6小鼠皮下,10天后收集海绵中的浸润淋巴细胞(SILs),经体外含rIL-2的培养系统中扩增后作特异性杀伤试验。结果表明:所获得的SILs对FBL-3瘤细胞显示出明显的特异杀伤活性,而且高于同一宿主脾脏中的致敏T细胞和未免疫鼠中SILs。经流式细胞仪分析表明,从免疫鼠中以的SILs是以Lyt2^+占绝对优势的T细胞系列,联合应用化疗与SI

Abstract:Tumor-specific T lymphocytes can be obtained from subcutaneous antigen-loaded sponge implants in mice. These cells were called sponge infiltrating lymphocytes (SILs)and cultured in presence of rIL-2 for expansion in vitro. The expanded SILs were transferred to tumor-bearing mice for therapy. The C57BL/6 mice were immunized with irradiated FBL-3 cells(a Friend virus induced leukemia)and then implanted with small piece of polyurethane sponges containing irradiated FBL-3 cells. The SILs were harvested on the tenth day,and expanded in culture with a low concentration of rIL-2. The specific cytolytic activity of SILs was tested by R-TdR release assay. The results demonstrated that SILs had specific cytolytic activity to FBL-3 cells which was greater than that of spleen cells from the same host and those SILs from the naive mice. Most of SILs were Lyt2 phenotype by FACS analysis. Adoptive transfer of the SILs in combination with cyclophosphamide (CY)showed a significant antitumor effect in tumor-bearing mice.
