参积护胃颗粒抗大鼠利血平型胃溃疡的实验研究

摘    要:目的通过观察参积护胃颗粒对利血平型大鼠胃黏膜损伤的保护作用,探讨参积护胃颗粒防治大鼠胃溃疡的作用。方法将60只健康SD大鼠分为对照组,模型对照组,奥美拉唑组,参积护胃颗粒低、中、高3个剂量组共六组,每组10只,雌雄各半,奥美拉唑组和参积护胃颗粒低、中、高剂量组的给药剂量分别为4.17mg/kg和2.61、5.22、10.44g/kg,其余两组予以等体积蒸馏水,灌胃7d,每天1次,末次灌胃后腹腔注射利血平(5mg/kg)复制胃溃疡模型,观察胃黏膜的损伤情况,计算溃疡指数和各组溃疡发生率,并取腺胃进行病理切片检测。结果与模型对照组比较.奥美拉唑组和参积护胃颗粒中、高剂量组均能显著降低胃溃疡指数,差异有统计学意义(P〈0.01)。奥美拉唑组的溃疡发生率为90.0%(9/10),参积护胃颗粒低、中、高剂量组的溃疡发生率分别为100.0%(10/10)、90.0%(9/10)、80.0%(8/10)。参积护胃颗粒中、高剂量组和奥美拉唑组的胃黏膜结构被破坏程度与模型对照组比较明显减轻。结论参积护胃颗粒对利血平所致的大鼠胃黏膜损伤具有保护作用。

Abstract:Objective Through the observation of Shenji huwei granules on reserpine-induced gastric mucosal lesion in rats, to discuss the role of Shenji huwei granules in prevention and treatmeat of rat' s gastric ulcer. Methods A total of 60 rats were divided into 6 groups with 10 cases in each group and sex in half:control group,model control group,omeprazole (OME) group, Shenji huwei granules group with low, middle and high dose. The OME group was treated with the dosage of 4.17 mg/kg, the Shenji huwei granules group with low, middle and high dose was administrated with the dosage of 2.61 g/kg, 5.22 g/kg and 10.44 g/kg respectively, and the other two groups were with isovolumetric distilled water. All the groups were intragastrically administrated for 7 days, once daily, and at the late of lavage, the model of gastric ulcer was copied by injecting 5 mg/kg reserpine intraperitoneally to observe the damage condition of gastric mucosa, calculate the ulcer index and the ulcer incidence rate of each group and con- duct the detection of pathological section. Results Compared with the model control group, OME group group and Shenji huwei granules group with middle and high dose could significantly reduce the ulcer index, and the difference was statistically significant (P〈0.01). The incidence rate of ulcer in OME group was 90.0% (9/10) ,while the occurrence rate of ulcer in Shenji huwei gran- ules group with low, middle and high dose was 100.0% (10/10), 90.0% (9/10) and 80.0% (8/10) respectively. Histopathological- ly, compared with the model control group,the damage degree of the gastric mucosal of Shenji huwei granules group with middle and high dose and OME group were significantly decreased. Conclusion Shenji huwei granules has a protective effect on reser- pine-induced rat's gastric mucosal.
