支气管哮喘大鼠淋巴液CD4+CD25+Treg及其Th1/Th2失衡的实验研究

摘    要:目的观察CD4+CD25+Foxp3+调节性T细胞、IL-4和IFN-γ在哮喘大鼠淋巴液与血液中的水平,探讨其与哮喘发病的关系。方法建立大鼠哮喘模型,收集激发后0、24、48 h淋巴液、血液中淋巴细胞,采用流式细胞术(FCM)检测淋巴液及血液中CD4+CD25+Foxp3+T细胞百分率;采用ELISA方法检测血浆及淋巴液中IL-4和IFN-γ水平。结果哮喘组淋巴液和血液的CD4+CD25+Foxp3+T细胞百分率水平、血浆IL-4和IFN-γ浓度在各时间点均低于对照组(P<0.05);哮喘组淋巴液中CD4+CD25+Foxp3+T细胞百分率水平、IL-4和IFN-γ浓度在不同时间点均高于血液水平(P<0.05)。结论哮喘大鼠淋巴液中存在CD4+CD25+Foxp3+调节性T细胞数量失调,且淋巴液中CD4+CD25+Foxp3+调节性T细胞水平显著高于其血液水平;哮喘大鼠淋巴液中存在以Th2亢进为特征的Th1/Th2失衡,淋巴液中细胞因子水平对于反映哮喘炎症程度可能更早和更准确。

Abstract:Objective To observe the expression of CD4+CD25+Foxp3+Treg、IL-4 and IFN-γ in lymph and blood,investigate the relationship between them.Methods Lymph and blood samples of 0 h,24 h,48 h after the last challenge were collected from the rat model of asthma.The percentage of CD4+CD25+Foxp3+Treg were detected by flow cytometer(FCM),while the levels of IL-4 and IFN-γ were determined by ELISA.Results The percentage of CD4+CD25+Foxp3+Treg in the blood and the levels of IL-4 and IFN-γ in the plasma in cases of asthma group were significantly lower than that of the control at different time points(P<0.05);The percentage of CD4+CD25+Foxp3+Treg and the level of IL-4 and IFN-γ in lymph were significantly higher than that of blood(P<0.05).Conclusion There is a disbalance both quantity of CD4+CD25+Foxp3+Treg in the lymph of bronchial asthmatic rat,and the percentage of CD4+CD25+Foxp3+Treg in lymph are significantly higher than that of blood.It is suggested that the Th1/Th2 unbalance which was impress of Th2 hyperfunction were existed in lymph of asthmatic rats.It could suppose that cytokines levels in lymph could reflex to the inflammation degree of asthma much early and accurately.
