乙肝病毒表面抗原基因疫苗诱导小鼠细胞及体液免疫应答的研究

摘    要:为观察乙肝病毒 H Bs Ag 基因疫苗免疫小鼠后的细胞及体液免疫应答,将该疫苗定向克隆于质粒pc D N A3 巨细胞病毒启动子下游,构建能在真核细胞内表达的重组基因疫苗pc D N A H Bs Ag 。将此疫苗通过多点肌肉注射免疫 Balb/ C 小鼠后第六周,实验组小鼠均出现抗- H Bs Ig G 阳性,其抗体水平明显高于对照组,同时 Th1 免疫应答相关的细胞因子 I L2 及γ干扰素水平也明显高于对照组,表明本文构建的基因疫苗pc D N A3 H Bs Ag 能诱导 Balb/ C 小鼠产生良好的细胞及体液免疫应答。

Specific cellular and humoral immune responses induced by intramuscular injection of DNA vaccine containing HBV HBsAg gene in mice
Abstract:To know the influences on the function of cellular and humoral immunity in mice by DNA vaccine containing HBV HBsAg gene, the DNA vaccine, pcDNA3 HBsAg, in which the HBsAg gene sequences of HBV were inserted into downstream of CMV promoter in pcDNA3, were injected intramuscularly into Balb/C mice. The results were that the serum anti HBsIgG was detected in all of the pcDNA3 HBsAg immunized mice, but none in control mice; serum levels of Th1 cytokines, interleukine 2, and interferon gamma, were significantly higher in pcDNA3 HBsAg immunized mice than these in controls. It is indicated that the good responses of cellular and humoral immunity can be induced by injection of pcDNA3 HBsAg in Balb/C mice.
