钉螺软体组织蛋白水解酶的初步分析

摘    要:目的分析钉螺软体组织中的蛋白水解酶。方法以明胶作底物,利用明胶-SDS-聚丙烯酰胺凝胶电泳(PAGE)分离钉螺软体组织蛋白质,于不同缓冲液和酶抑制剂中进行孵育,分析和鉴定其中的蛋白水解酶。结果钉螺软体组织中存在降解明胶的蛋白水解酶,其最适pH值为碱性。金属蛋白酶抑制剂乙二胺四乙酸二钠(EDTA)可抑制其活性。电泳分离获得有活性的酶蛋白。结论钉螺软体组织中存在被EDTA抑制的蛋白水解酶。

Abstract:Objective To analyze the proteinase in soft tissues of Oncomelania hupensis.Methods The proteinases of Oncomelania hupensis snails were fractionated through gelatin-SDS-PAGE and the gelatin was degraded by proteinases when the gel was incubated in buffer with optional pH.The metalloproteinase that degraded the gelatin was identified by incubating gel with optional buffer containing different enzyme inhibitors.The metalloproteinase was isolated by SDS-PAGE.Results There was a metalloproteinase in the extracts of the snails.The optional pH of metalloproteinase was 9.The metalloproteinase was inhibited by EDTA.The metalloproteinase demonstrated the enzymatic activity after being fractionated through SDS-PAGE.Conclusion A proteinase inhibited by EDTA exits in the extracts of soft tissue of Oncomelania hupensis.
