摘    要:目的探讨食管粘膜BAT-26位点MSI及其单态性的情况.方法组织DNA提取后PCR扩增,扩增产物行9%变性聚丙烯酰胺凝胶电泳,最后银染等方法, 分析45例食管癌及正常切缘组织BAT-26位点微卫星不稳的情况.结果所有正常组织在BAT-26位点DNA电泳条带长度无明显改变,45例食管癌中有3例MSI改变.结论食管粘膜BAT-26具有较好的单态性,食管癌细胞BAT-26位点对识别高频率MSI不十分敏感.

Abstract:Objective To investigate the state of microsatellite instability and monomorphism at microsatellite DNA BAT 26 loci of esophageal mucosa.Methods Genomic DNA extracted from tissues was PCR amplified.PCR products were run on 9% denaturing polyacrylamidegel and stained with silver.By using these methods,we analysised MSI and monomorphism of microsatellite BAT 26 loci at normal tissue and esophageal cancer tissue.Results All normal specimens showed no variation in the length of MS DNA at BAT 26 loci.MSI were detected in 3 of 45(6.7%)esophageal cancer analyzed.Conclusion BAT 26 has much monomorphism in genome of normal esophageal tissue.BAT 26 loci of esophageal cancer is not sensible in identifying MSI.
