慢性氟中毒对大鼠心肌胶原代谢的影响

摘    要:目的:探讨过量氟对大鼠心肌组织Ⅰ型、Ⅲ型胶原蛋白表达的影响。方法:将Wistar大鼠随机分成5组(每组8只):常规食对照组、常规食加氟组(加氟100 mg·L-1)、低钙偏食对照组、低钙偏食加低量氟组(加氟50 mg·L-1)和低钙偏食加高量氟组(加氟100 mg·L-1)。采用苦味酸-天狼星红染色、偏振光显微镜观察以及免疫组织化学法检测氟中毒大鼠心肌Ⅰ型、Ⅲ型胶原的表达。结果:图像分析显示苦味酸-天狼星红染色、偏振光显微镜下显示的Ⅰ型、Ⅲ型胶原面积百分比,各加氟组其Ⅰ型、Ⅲ型胶原均较其对照组稍有增高趋势,但差异无显著性(P>0.05)。免疫组化观察Ⅰ型、Ⅲ型胶原蛋白的表达量0%~10%,各加氟组与其对照组相比差异亦无显著性(P>0.05)。结论:慢性氟中毒对大鼠心肌胶原代谢无明显影响,心肌胶原可能不是氟化物作用的靶子。

Abstract:Objective To study the effect of fluoride on the expressions of collagen type I (Col I ) and collagen type III (Col III ) in the myocardium of rats. Methods The rat model was established by administration of different concentration of sodium fluoride. The myocardium of rats were stained by picrosirium red, observed by polarization microscopy, analyzed by image analysis system. The expressions of Col I and Col III in myocardium of rats were determined by immunohistochemistry method. Results The percentages of Col I and Col III area in the myocardium of rats treated with different concentration of sodium fluoride were a little higher than those of control group. No significant difference was seen among different groups of the expressions of Col I and Col III (P>0. 05). The expressions of Col I and Col III protein were ranged between 0 - 10%. There was no significant difference among different groups either (P>0. 05). Conclusion Chronic fluorosis has no obvious effect on myocardial collagen metabolism of rats and myocardial collagen is not likely the main target of fluoride.
