摘    要:目的整体培养包括上下软骨终板、纤维环及髓核组织的大鼠椎间盘器官,观察髓核组织的变化,探索一种椎间盘器官整体培养的实用技术。方法取SD大鼠90只,完整取出腰段椎间盘器官共400个,随机分为4组,置入12孔培养板内,配制高渗培养液,对椎间盘器官进行整体培养,观察髓核细胞成活率、组织完整性及基质变化情况。结果体外培养2周的椎间盘组织形态、细胞存活率及髓核组织成分与新鲜离体椎间盘无统计学差异。结论利用高渗培养液可对椎间盘器官进行整体培养,为椎间盘的生理与病理的研究提供一种理想的模型。

The changes of the nucleus pulposus after the whole intervertebral discs organ culture

Abstract:Objective To find a practical method of culturing discs organ system by observing the changes of the nucleus pulposus after the whole intervertebral discs,including cartilage eng-plate nucleus,pulposus and anulus fibrosus,were cultivated.Methods 400 intervertebral discs were taken out completely from 90 SD rats and put to the culture plate after being divided into 4 groups randomly.Cultivate the whole intervertebral discs with high osmotic culture medium,then observe the cell viability,tissue integrity and morphology of them.Result The cell viability,tissue integrity and morphology of the whole intervertebral discs were not changed statistically.Conclusion Our system could be used to cultivate the whole intervertebral discs for next investigations.
