脑伤泰对血管性痴呆大鼠P300与海马突触素的影响

摘    要:目的探讨血管性痴呆大鼠P300与海马突触素(synaptophysin,SYN)在活血化瘀中药脑伤泰作用下的变化,并探讨其机制。方法选用3月龄,雄性Wistar大鼠,体质量(200±10)g,由第三军医大学实验动物中心提供。采用随机数字表分为假手术组、模型组与治疗组,每组37只,其中行为学训练每组7只,P300测定每组5只,免疫组化测定每个时象点5只。采用Pulsinlli-4血管法制作血管性痴呆大鼠模型,分为假手术组、模型组、治疗组,采用主动回避反应测定行为学变化,脑诱发电位仪测定P300潜伏期,免疫组化法行海马突触素染色并进行图像分析测定其光密度值统计分析。结果模型组主动回避反应正确率明显下降,P300潜伏期明显延长、海马SYN光密度值明显下降,治疗组主动回避反应正确率明显下降,P300延长,海马SYN光密度值明显下降,但在术后8周主动回避反应正确率明显恢复,P300缩短,海马SYN光密度值明显上升。结论血管性痴呆大鼠主动回避反应正确率明显下降,P300潜伏期明显延长,海马SYN水平明显下降,脑伤泰提高VD大鼠主动回避反应正确率,缩短P300潜伏期,其机制可能与提高海马突触素水平有关。

Abstract:Aim To investigate the changes of P300 and synaptophysin in hippocampus of vascular dementia rats under the effect of Naoshangtai( NST) ,a tranditional Chinese medicine mixture and explore its mechanism.Methods 3-month old male Wistar rats weighed ( 200± 10)g were provided by experiment animal center,the Third Military Medical University and divided by random number table into false operation group,model group and treatment group with 37 rats in each group. In each group,7 rats were used for praxiology training,5 rats for P300 determination and 5 rats for immunohistochemical determination. The rat model of vascular dementia was established by Pulsinlli-4-vesselocclusion method.Active avoidance reaction was used for determination of praxiological changes,cerebral evoked potential device for latent period of P300.Hippocampus synaptophysin was stained immunohistochemically and optic density was measured by image analysis.Results The correct rate of active avoidance reaction was decreased obviously,latent period of P300 was prolonged obviously and optic density of hippocampus SYN decreased obvious in model group and treatment group.But,at 8 week after operation,correct rate of active avoidance reaction was obviously resumed,P300 shortened and optic density of hippocampus SYN increased in treatment group.Conclusion Naoshangtai can increase the correct rate of active avoidance reaction,shorten latent period of P300 and the mechanism might be related to increasing of hippocampus SYN level.
