高效液相色谱法测定双黄连粉针中黄芩甙和绿原酸的含量

本文采用高效液相色谱法，测定中药制剂双黄连粉针中黄芩甙和绿原酸含量。流动相分别是0．1mol／L磷酸二氢钾：乙酸：四氢呋喃（100：10：19）和甲醇-10％磷酸缓冲液（10：90），流速分别是0．8和1ml／min；色谱柱ERC-ODS-1161（6×100mm），柱温55℃；黄芩甙以电化学为检测器，检测电压是＋650mv；绿原酸以紫外为检测器，检测波长274nm；结果：该方法的最小检测浓度分别为0．025mg／L和0．1mg／L，平均回收率分别为100．1％和100．4％，重现性好。本方法测定快速、简便、准确，适用于含黄芩甙和绿原酸的中药制剂的含量测定。

Determination of the Contents of Baicaline and Chlorogenic Acid in Shuanghuanglian Powdery Injection by Method of HPLC

The contents of baicaline and chlorogenic acid in Shuanghuanglian powdery injection have been determined by themethod of HPLC under following conditions:column-ERC-ODS-1161,6 × 100mm;temperature 55℃;flow phase of baicaline-0.1 mol/LKHPO,acetic acid and tetrahydrofuran(1000：100:190);flow phase of Chlorogenic acid-methanol and 10%phosphate-buffer(10:90);flowrate 0.8ml/min;volt of ECD-detecor of baicalinee-＋650mv;wavelength of UV-detector of chlorogenic acid 274nm.The resultsshowed that the minimun determinable concentrations of baicaline and chlorogenic acid were 0.025mg/L and 0.1mg/Lrespectively,with an average recovery of 100.1% and 100.4% respectively.This method is highly reproducible,rapid,simple,convenient,accurate and suitable for the content determination of baicaline and chlorogenic acid in Chinese medicinal preparations.
