实验性脑出血大鼠脑组织血管内皮生长因子的表达及意义

摘    要:目的 探讨实验性脑出血大鼠脑组织血管内皮生长因子(VEGF)的表达及其意义。方法 应用立体定向技术于大鼠尾壳核注射自体尾动脉血建立大鼠脑出血模型,动态观察组织病理学改变,动态观察脑组织含水量。免疫组织化学染色显示脑出血后VEGF在脑组织内的表达。结果 大鼠脑出血后脑含水量于24h开始明显升高。48h达高峰并持续到72h,与病灶对侧及假手术组比较有统计学意义;脑出血后脑内VEGF阳性细胞数随时间逐渐增多。以24h,48h,72h最为显著,7d时呈现血管样结构。结论 脑出血后脑内VEGF的表达与脑水肿相关,且VEGF可促进血肿周围组织的血管增生,可能对脑出血后的组织修复起促进作用。

Abstract:Objective To investigate the Expression and Significance of Vascular Endothelial Growth Factor in Rats after Experimental Intracerebral Hemorrhage (ICH).Methods With stereotxic technique,Rat model of ICH was established by injection of autologous caudal artery blood into the rat caudate nucleus.The pathology observed dynamically,brain water content was measured,and immunohistochemical methods were performed to examine the expression of VEGF in rat brain at 6 h,12 h,24 h,48 h,72 h,7 d respectively.Results After ICH,the amount of water in rat brain began to go up at 24h,reached maximum at 48h and continued to 72 h.Compared with the shamp group,there was significant difference.The amount of VEGF positive cells increased evidently with the time going up,at 24h,48h and 72h,it was obvious,and at 7d,it showed the blood vessel formation.Conclusion The expression of VEGF in brain after ICH is relative to brain edema.VEGF can accelerate the hyperplasia of blood vessel round hematoma,and have positive effect on tissue repair after ICH.
