摘    要: 目的在α-酸性糖蛋白手性柱和纤维素-三(4-甲基苯甲酸酯)手性柱上,对两种四氢原小檗碱同类物(延胡索乙素和四氢小檗碱)进行了对映体分离研究。方法在α-酸性糖蛋白手性柱上考察了流动相的有机改性剂、pH值以及离子强度对分离的影响,在纤维素-三(4-甲基苯甲酸酯)手性柱上主要考察了流动相体系、流速和柱温对分离的影响,并初步探讨了手性识别机制。结果延胡索乙素在两种手性柱上均达到基线分离,四氢小檗碱只在纤维素-三(4-甲基苯甲酸酯)柱上分离成两个成分。结论延胡索乙素和四氢小檗碱均可分离成两个成分,推断为其对映体的拆分,可为进一步探讨其在体内的动态变化提供方法学基础。

Abstract:OBJECTIVE To study the enantiomeric separation of two tetrahydroprotoberines(dl-tetrahydropalmatine and dl-tetrahydrobeberine) on two chiral columns based on alpha acid glycoprotein(AGP) and cellulose tris(4-methylbenzoate) by HPLC.METHODS The effects of pH,buffer concentration and organic modifier on the chiral resolution with the AGP column were investigated.The effects of mobile phase and column temperature were also studied using the cellulose derivative column.RESULTS A baseline separation of THP enantiomers was obtained on both columns,while THB enantiomers were separated on the cellulose column.CONCLUSION The enantioseparation method can be applied for the further investigation of THP and THB enantiomers in vivo.
