摘    要:目的探讨慢性压力负荷对兔左心室心肌细胞凋亡的影响。方法采用末端脱氧核苷酸转换酶介导的dUTP-生物素平移末端标记技术,观察持续压力超负荷引起的心力衰竭兔的左心室心肌细胞凋亡的动态变化。结果对照组仅有极少量心肌细胞凋亡,实验组凋亡细胞水平迅速增加,术后第1天已与对照组有显著性差异(P<0.001),术后第3天与术后第7天之间存在一个高峰值,术后第7天后凋亡细胞水平渐下降,术后第14天已下降至术后第1天的水平,而在心衰临床症状出现时凋亡细胞水平复又明显高于其他各组(P<0.001)。结论正常机体在慢性压力超负荷作用下,早期即出现凋亡细胞增加,并存在一波浪式动态变化,其峰值出现时间早于心肌显著肥厚时间。

Abstract:Objective To studytheeffectof chronicpressureoverloadon theapoptosisof theleftventriclemyocytesin rabbits.Methods Rabbitmodelsof chronicpressureoverload-inducedheartfailurewerepreparedinwhichdynamicchanges of apoptoticmyocytesin theleftventriclewereobservedby way of terminaldeoxynucleotidyltransferase-mediateddUTP nickend-labeling(TUNEL)assay.Results Onlya fewapoptoticcellswas observedinthesham-operatedgroup,whileinthe experimentalgroup,theapoptoticleftventriclemyocytessignificantlyincreasedafteroperation,presentinga peaklevel betweenday3andday7.Sevendaysaftertheoperation,theapoptoticmyocytesbeganto decreaseandtillday14,the apoptoticcellnumberhadbeensmallerthanthatmeasuredon day1.Whensignsof heartfailuresetin,theapoptoticmyocytes wereagainincreased(P<0.001).Conclusion Duringchronicpressureoverload,myocyteapoptosisintheleftventricleis elevatedattheearlystagesandundulatessubsequently,withthepeakoccurringbeforehypertrophyisobvious.
