摘    要:目的选择黄芪药材高效液相色谱-二极管阵列检测法(HPLC-DAD)指纹图谱的最佳检测波长。方法提取可用于评价指纹图谱质量的相关信息数据,借助化学计量学的方法对峰数、分离度之和、相对峰面积之和、互信息ФA和互信息ФB等指标参数进行比较和分析。结果在260 nm波长下峰数、分离度之和、相对峰面积之和、互信息ФA和互信息ФB参数值分别为30(17)、190.43、4.19、2.48、737.23,均优于249 nm和280 nm波长条件下相应的参数值。结论从化学计量学方法确定的指标参数结果确定260 nm为黄芪中黄酮类成分指纹图谱的最佳检测波长,可以为HPLC-DAD指纹图谱最佳检测波长的选择提供可靠依据。

Abstract:Objective To select and confirm the optimized detection wavelength of the fingerprints of Radix Astragali obtained by HPLC-DAD.Methods The related information to evaluate the intrinsic quality of fingerprints was extracted and processed with methods of chemometrics to the peak numbers,sum of resolution,sum of relative peak area,mutual information ФA and mutual information ФB,by which the index parameters and functions were established.Results At the wavelength of 260 nm,the index parameters and functions of the peak numbers,sum of resolution,sum of relative peak area,mutual information ФA and mutual information ФB were 30(17),190.43,4.19,2.48 and 737.23,better than the index parameters of 249 nm and 280 nm.Conclusion From the results of the index parameters established by chemometrics,260 nm can be confirmed as the best detection wavelength of fingerprints of flavonoids in Radix Astragali,and can provide credible evidence to select the optimized detection wavelength of the fingerprints for HPLC-DAD.
