P-选择素对角膜上皮创伤修复及其中性粒细胞迁移的影响

摘    要:目的观察P-选择素对角膜上皮创伤修复及其中性粒细胞迁移的影响。方法选用P-选择素基因敲除小鼠与野生型小鼠进行对比,观察角膜上皮创伤修复过程,同时用免疫组织化学方法观察中性粒细胞在角膜的迁移,并用酶联免疫吸附测定方法检测角膜中细胞因子和趋化因子的变化。结果与野生型小鼠相比,P-选择素基因敲除小鼠的角膜创伤修复延迟,向角膜迁移的中性粒细胞数量减少;中性粒细胞的迁移与趋化因子和细胞因子有密切的关系。结论P-选择素表达缺陷可导致角膜上皮创伤修复的延迟。这种延迟可能与创伤后中性粒细胞向创伤区迁移数量减少有关。

Impacts of P-selectin on corneal epithelial wound healing and neutrophil migration into wound

Abstract:ObjectiveTo observe the impact of P-selectin on corneal epithelial wound healing and neutrophil migration into wounded corneas.MethodsCorneal wound model was established by scaling the corneal epithelium under the microscope in P-selectin gene knockout mice and wild type mice.The corneal epithelial wound healing procedure was observed during the 48 hours after wound at a 6 hours interval by fluorescein corneal staining.The number of neutrophils migrating into the wound at different time points were counted.The changes of chemokines and cytokines and interleukin-6 were evaluated in the corneas using ELISA.Results The corneal epithelial wound healing took about 24-38 hours in P-selectin gene knockout mice but it was 48 hours in wild type mice,showing a significant delay in the P-selectin gene knockout mice (P<0.05).Migration of neutrophil into the wound exhibited the first peak at 6-24 hours and the second peak at 30-48 hours in wild type mice,but that of P-selectin gene knockout mice was 24 hours and the number of neutrophil was significantly decreased in P-selectin gene knockout mice compared with wild type mice(P<0.05).Chemokines,cytokines changes appeared to be a similar pattern to neutrophil migration.Conclusion The delay of corneal epithelial wound healing in the P-selectin gene knockout mice may be due to the decrease of neutrophil migration into the wounded corneas.There were a close relationship among chemokines,cytokines and neutrophil migration.
