红景天苷对人肝癌HepG2细胞增殖及凋亡的影响

摘    要: 目的 观察红景天苷(salidroside)对人肝癌HepG2细胞增殖和凋亡的影响, 探讨其抗肿瘤的可能机制。方法 以体外培养的人肝癌HepG2细胞作为研究对象, 倒置显微镜观察红景天苷对肝癌细胞形态的影响;采用MTS法检测红景天苷对细胞生长的抑制作用;采用流式细胞仪检测细胞凋亡率;Western blot法检测Bcl-2、Bax、caspase-3、caspase-8的表达。结果 红景天苷能显著抑制肝癌细胞HepG2增殖并促进其凋亡, 下调细胞Bcl-2 表达, 上调Bax、caspase-3及caspase-8的蛋白表达量。结论 红景天苷在体外对人肝癌HepG2细胞具有显著的抑制增殖和促进凋亡作用, 其机制可能与其调节凋亡基因Bcl-2、Bax、caspase-3与caspase-8的蛋白表达有关。

Abstract:OBJECTIVE To investigate the salidroside on proliferation and apoptosis of human hepatocellular carcinoma HepG2 cells and its possible mechanisms of anti-tumor. METHODS Cultured HepG2 cells in vitro were used as the research object and their cell morphology were observed by inverted microscope. The inhibitory rate of cell proliferation was measured by MTS assay. The cell apoptosis was determined by flow cytometry.The protein expressions of Bcl-2, Bax, caspase-3 and caspase-8 were detected by Western blot analysis.RESULTS Salidroside could inhibit the proliferation of HepG2 cells and promote its apoptosis. The Bax, caspase-3 and caspase-8 protein expression were increased while Bcl-2 protein expression was decreased in HepG2 cells in a concentration-dependent manner. CONCLUSION Salidroside can inhibit the proliferation and induce the apoptosis of HepG2 cells, and the mechanism may be associated with regulating protein expression of Bcl-2, Bax, caspase-3 and caspase-8.
