肝X受体:糖和脂代谢中的重要调节者

摘    要:肝X受体(LXRs)是核受体超家族成员,能被氧化的胆固醇衍生物结合并激活,在胆固醇逆向转运中起着非常重要的作用。在肝细胞中,胰岛素能诱导LXRα表达,抑制糖异生关键酶包括磷酸烯醇式丙酮酸羧激酶(PEPCK)、葡萄糖-6-磷酸酶(G-6-Pase)、果糖1,6-2磷酸酶(FBP1)],故蹦凡可作为糖尿病治疗中重要的药物作用靶点。另外,LXRα还可激活SREBP1c的表达,从而在脂肪酸代谢中也发挥重要作用。本文简要介绍了LXRs凡在糖代谢和脂代谢中的分子作用机制。

Abstract:Liver X receptors (LXRs) are members of the nuclear receptor superfamily and are activated by oxysterols and intermediates in the cholesterol synthetic pathway. The pivotal role of LXRs in the metabolic conversion of cholesterol to bile acids has been well established. Furthermore, insulin induces LXRa in hepatocytes, resulting in the suppression of key enzymes in gluconeogenesis, including phosphoenolpyruvate carboxykinase, glucose-6-phosphatase, and fructose-1, 6-bisphosphatase (FBPase). LXRs also play an important role in fatty acid metabolism by activating the sterol regulatory element-bing protein 1c gene (SREBP1c). This articles reviews the molecular mechanisms by which LXRs act to influence the lipid and carbohydrate metabolism.
