子宫内膜癌组织中 GDF -15、p -mTOR 蛋白的表达及相关性

摘    要:目的:研究 GDF -15和 p -mTOR 蛋白在子宫内膜癌组织中的表达及其与临床生物学特点的相关性,并分析恶性肿瘤发生发展和转移的内在信号通路。方法:免疫组织化学方法检测 GDF -15和 p -mTOR蛋白在65例子宫内膜癌和65例正常子宫内膜组织中的表达。结果:GDF -15和 p -mTOR 蛋白在子宫内膜癌组织中的表达率均明显高于对照组,差异有显著性意义(P <0.01)。GDF -15蛋白在Ⅰ-Ⅱ期子宫内膜癌组织中的表达率为52.94%,Ⅲ-Ⅳ期中的表达率为90.32%,两者比较有显著性差异(P <0.01)。GDF -15在癌灶肌层浸润深度≥1/2中表达率为92.00%,在癌灶肌层浸润深度<1/2组的表达率为57.50%(P <0.01)。GDF -15在存在肿瘤淋巴结转移的表达率为85.71%,无淋巴结转移的表达率为59.46%,两者相比有显著性差异(P =0.02)。p -mTOR 蛋白在Ⅰ-Ⅱ期子宫内膜癌组织中的表达率为64.71%,Ⅲ-Ⅳ期中的表达率为93.55%,两者比较有显著性差异(P <0.01);p -mTOR 在癌灶肌层浸润深度≥1/2中表达率为92.00%,在癌灶肌层浸润深度<1/2组的表达率为70.00%,两者相比有显著性差异(P <0.05)。p -mTOR 在存在肿瘤淋巴结转移的表达率为92.86%,无淋巴结转移的表达率为67.57%,两者相比有显著性差异(P <0.05)。GDF -15、p -mTOR 的表达与患者的发病年龄、组织病理学分级无关(P >0.05)。子宫内膜癌组织中 GDF -15和 mTOR 蛋白表达呈显著正相关(r =0.322,P <0.01)。结论:p -mTOR 在子宫内膜癌中的表达受升高的 GDF -15蛋白水平的调控,可能为子宫内膜癌发生发展及侵袭转移的重要信号转导通路。

Expression and correlation between GDF -15 and p -mTOR in endometrial carcinoma
Abstract:Objective:To study the expression of GDF -15 and p -mTOR protein and the relationship with the pathogenesis and development of endometrial cancer so as to elucidate the role of mTOR signal transductive pathway. Methods:The expression of GDF -15 and p -mTOR protein was detected in 65 endometrial cancer and 65 normal endometrial tissues with immunohistochemistry.Results:The positive expression rates of GDF -15 and p -mTOR protein in endometrial carcinoma tissue were significantly higher than those in the normal endometrial tissue one's re-spectively(P <0.01).The positive rates of GDF -15 and p -mTOR protein were significantly higher in endometrial carcinoma with muscle invasive depth 1 /2 muscular layer than in that <1 /2 muscular layer(92.00% vs 57.50%, 92.00%vs 70.00% respectively),higher in Ⅲ or higher FIGO stages than in Ⅰor Ⅱstages (90.32% vs 52.94%, 93.55% vs 64.71% respectively),higher in patients with lymph node metastasis than in those without(85.71% vs 59.46%,92.86% vs 67.57% respectively).The positive expression of GDF -15 and p -mTOR was not associated with age or differentiation degree(P >0.05).GDF -15 positive cases were significantly positively correlated with p -mTOR positive cases(r =0.322,P <0.01).Conclusion:GDF -15 can promote p -mTOR in the process of patho-genesis and development in endometrial carcinoma and they maybe the important signal transduction pathway.
