腺苷抑制乳腺癌细胞增殖和迁移的机制

目的:检测腺苷对乳腺肿瘤细胞增殖及迁移能力的影响，并探究其潜在的分子机制。方法:以MCF-7细胞为模型，采用梯度浓度的腺苷处理细胞，MTT及细胞计数法检测腺苷对细胞增殖的影响，流式细胞术检测腺苷对细胞凋亡及周期的影响，划痕修复实验检测腺苷对细胞迁移能力的影响。结果:腺苷可显著抑制MCF-7细胞增殖，且抑制效果与腺苷浓度及作用时间呈相关性。流式细胞术检测结果表明腺苷可诱导MCF-7细胞凋亡，并引起细胞的 G2/M期阻滞。划痕实验结果表明腺苷抑制MCF-7细胞的迁移能力。实时定量PCR检测结果发现，促细胞凋亡分子Bax及Bak表达量显著升高，而凋亡抑制分子Bcl-2表达量显著下降，同时Caspase-3的表达量也显著升高。结论:腺苷可诱导MCF-7细胞凋亡，抑制细胞的增殖，同时腺苷可以抑制MCF-7细胞的迁移能力。腺苷诱导MCF-7细胞凋亡与其激活线粒体凋亡通路密切相关。

Adenosine inhibits proliferation and migration of breast cancer cells

Objective:To evaluate the effect of adenosine on breast tumor cell proliferation and migration,and ex-plore the potential molecular mechanisms.Methods:The effect of adenosine on MCF -7 cells proliferation was detec-ted by MTT and cell counting assay.Cell apoptosis and cell cycle was examined using FACS calibur flow cytometer. And the effect of extracellular adenosine on migration of MCF -7 cells was examined by scratch assay.Results:Aden-osine significantly suppressed MCF -7 cells proliferation in a dose -and time -dependent manner.The flow cytome-try analysis showed that adenosine could also induce breast cancer cell apoptosis.Moreover,adenosine treatment also decreased the migration of MCF -7 cells.Real -time quantitative PCR results showed that pro -apoptotic molecules were up -regulated while anti -apoptotic molecule was decreased in response to adenosine treatment.Conclusion:Adenosine can suppress breast cancer cells proliferation,migration and induction of apoptosis via mitochondrial apop-tosis pathway.
