摘    要:目的:通过体外细胞观察多发性骨髓瘤患者本周氏蛋白酰胺酶催化作用与肾小管细胞毒性作用关系,进一步了解本周氏蛋白对多发骨髓瘤肾损伤毒性机制。方法:选取22例多发性骨髓瘤患者为研究对象,以不同浓度的本周氏蛋白和猪肾小管上皮细胞共同培养1天后,用四甲基偶氮唑盐检测细胞增殖抑制率,以抑制细胞增殖的本周氏蛋白和猪肾小管上皮细胞共同培养,使用流式细胞术检测细胞凋亡。结果:肾损伤多发性骨髓瘤较无肾损伤更易对猪肾小管上皮细胞产生毒性作用,且本周氏蛋白催化常数值越高,则细胞毒性越强。本周氏蛋白浓度越高,猪肾小管上皮细胞凋亡和坏死率越高,为正相关,5mg/ml、10mg/ml和0mg/ml比较差异显著(P<0.05)。结论:本周氏蛋白酰胺酶催化作用强度和毒性作用正相关,可能是多发骨髓瘤患者肾损伤机制之一。

Abstract:Objective:To observe patients with multiple myeloma Bence Jones protein lactamase catalysis and re-nal tubular cells in vitro cytotoxicity relationships.Methods:Eleren cases of multiple Bence Jones protein lactamase Michaelis constant kinetic parameters and catalytic constant urine myeloma patients,with different concentrations of Bence Jones proteins and porcine renal tubular epithelial cells one day after co -culture with tetrazolium salts detect cell proliferation rate.Results:Multiple myeloma kidney damage was more than those without renal tubular epithelial cells of the pig produce toxic effects,and Bence Jones proteins catalyze a constant value was higher.Conclusion:Cat-alytic protein -lactamase was positively correlated with the intensity and toxicity.
