基质金属蛋白酶基因多态性与食管癌易感性的 Meta 分析

摘    要:目的:探讨 MMP 基因多态性与食管癌易感性之间的关系。方法:使用 Pubmed、Web of science、Em-base、中国生物医学文献数据库、中国知网、万方数据库检索2015年5月以前 MMP1-16071G/2G、MMP2-1306 C >T、MMP9-1562 C >T 与食管癌易感性的相关病例-对照研究,采用 STATA 12.0软件进行 Meta 分析。结果:共纳入10篇文献,3159例食管癌患者和4405名正常对照者。MMP1-16071G/2G 多态性与高加索人食管癌的发生相关,与1G 等位基因相比,2 G 等位基因显著增加食管癌的患病风险(2G vs 1G,OR =1.38,95%CI =1.18~1.61,P <0.05)。但在亚洲人群的亚组分析中,未发现有统计学意义。MMP2-1306 C>T 基因多态性中,相对于 C 等位基因,T 等位基因是食管癌的保护因素(OR =0.75,95%CI =0.65~0.85,P<0.05)。而 MMP9-1562 C >T 多态性并没有影响食管癌的发生。结论:MMP1和 MMP2基因多态性可能与食管癌易感性相关,尚不能排除 MMP9在食管癌发病中的作用,仍需进一步研究。

Meta analysis of association between polymorphisms of MMP gene and risk of esophage-al cancer
Abstract:Objective:To explore the contribution of metalloproteinase matrix(MMP)gene polymorphisms to sus-ceptibility to esophageal cancer.Methods:The case -control studies involving the association of MMP1 -1607 1G/2G,MMP2 -1306 C >T and MMP9 -1562 C >T polymorphism with the susceptibility to esophageal cancer were re-trieved through computer -based search in PubMed,Web of science,Embase,Chinese Biomedical Literature Data, China National Knowledge Infrastructure and Wanfang Database before May,2015.The statistical analysis was per-formed by STATA 12.0 software.Results:A total of ten eligible case -control studies,including 3 159 esophageal cancer cases and 4 405 controls,were included for analysis.MMP1 -1607 1G/2G polymorphism was significantly cor-related with the Caucasian.Compared with 1G allele,the 2G allele polymorphism increased esophageal cancer risk(2G vs 1G,OR =1.38,95%CI =1.18 ~1.61,P <0.05),but in the Asian population subgroup analysis,the statistical significance was not found.For MMP2 -1306 C >T gene polymorphism,compared with the C allele,T allele carriers suffering from esophageal cancer risk decreased(T vs C OR =0.75,95%CI =0.65 ~0.85,P <0.05),and MMP9 -1562 C >T polymorphism did not influence the occurrence of cancer of the esophagus.Conclusion:Polymorphism of MMP1 and MMP2 may be related to the susceptibility of esophageal cancer,and it is not possible to rule out the sig-nificant association of MMP9 with esophageal cancer.
