胆碱对酒精中毒孕鼠神经元保护作用的实验研究

目的:观察不同剂量胆碱对孕期酒精中毒大鼠生仔后神经元的影响,探讨其机制。方法:将孕鼠分为酒精组、大剂量胆碱组、小剂量胆碱组及对照组。用灌胃法建立酒精中毒动物模型。在分娩当天进行行为测试,分别观察母鼠的舔仔行为和哺乳行为,计算各行为时间,比较各组母鼠的行为的差异。用免疫组化方法检测下丘脑室旁核催产素神经元数量变化。结果:酒精组母鼠的行为持续时间与对照组比较差异有显著性(P<0.01或P<0.05);胆碱组的行为持续时间与酒精组比较差异有显著性(P<0.01或P<0.05);与对照组比较,酒精组和小剂量胆碱保护组PVN区催产素神经元阳性细胞数目减少(P<0.05)。结论:怀孕期大量饮酒能够明显损伤相关的催产素神经元,而胆碱对神经元有一定的保护作用,并具有一定的剂量依从性,其机制可能是足量的胆碱为脑细胞提供了一定的营养,有效的保持了细胞结构。

The protective effect of chloride choline on the neuron of pregnant rats with alcoholism

Objective:To observe the effect of chloride choline on the behavior of alcoholism rat and to study its mechanisms.Methods:The pregnancy rats were randomly divided into alcoholism group,high dosage choline group,low dosage choline group and control group.The model were set up by intragastric administration.After delivery,behaviors(arched-back nursing,licking) were monitored by VHS to detect the amount of oxytocinergic neuron in paraventricular nucleus of hypothalamus.Results: The behavior in model groups changed comparing to that in control group(P<0.01 or P<0.05);that in high dosage choline group and lowdosage choline group changed comparing with that in alcoholism group(P<0.01 or P<0.05);The amount of oxytocinergic neuron of paraventricular nucleus of hypothalamus in alcoholism group and low dosage choline group were lower than that of the control group(P<0.05).Conclusion:Alcoholism in pregnancy can impair the behavior,and the choline can protect the behavior.
