组胺和甲氰咪胍对粒-单系祖细胞的影响

摘    要:用体外琼脂培养法研究组胺和甲氰咪胍对小鼠粒-单系祖细胞的影响,结果表明,10~(-9)M~10~(-5)M 组胺和10~(-8)M~10~(-4)M 甲氰咪胍分别作用时,对粒-单系祖细胞的产率没有影响,用甲氰咪胍阻断 H_2-受体后,组胺可增加粒-单系祖细胞的产率和自杀率。对此结果的理论意义和临床意义进行了讨论。

Abstract:The effects of histamine and cimetidine on the proliferation of granulocyte-macrophage progenitor(CFU-GM)of LACA mouse bone marrow was investigatedby using the agar culture technique.It was found that histamine alone,atconcentration of 10~(-9)M 10~(-5)M,and cimetidine alone,at concentration of 10~(-8)M10~(-4)M,had no effects on the granulocytemacrophage colony formation.When thehistamine H_2-receptor was blocked by cimetidine(10~(-5)M),histamine treatment(10~(-8)M 10~(-5)M)resulted in a significant increase of colony number as comparedto the untreated controls(P<0.001).The treatment in vitro of mouse bonemarrow cells with cimetidine(10~(-5)M)and histamine(10~(-6)M)increased thehydroxyurea suicide rate of CFU-GM from 27.7% to 43.0%.These resultssuggested that both histamine H_1-and H_2-receptor were present on thegranulocyte-macrophage progenitor.The stimulating H_1-receptor enhanced thegranulocyte-macrophage colony formation,while the effect of H_2-receptorsantagnited the effect of H_1 receptor.
