Abstract:Objective:To provide theoretical basis for the diagnosis and targeted therapy of pancreatic ductal adenocarcinoma(PDAC),we excavated the hub genes of PDAC and investigated the expression and prognostic value via bioinformatics analysis.Methods:Gene chip data GSE16515,GSE28735,GSE62452 and TCGA-GTEx were downloaded from GEO and TCGA databases,respectively,then we found common differential expressed genes through R software.GO enrichment analysis and KEGG pathway analysis were conducted,and protein-protein interaction(PPI)Network was constructed to screen hub genes,and finally we verified the expression of hub genes in PDAC and their prognostic value of PDAC.Results:A total of 219 DEGs were identified,of which 139 were up-regulated and 80 were down-regulated.Finally,a total of 10 key genes associated PDAC were screened,among which FN1,POSTN,VCAN,MMP1,EGF,and ALB were significantly related to the poor prognosis of PDAC(P<0.05).VCAN combined with ALB,MMP1 and POSTN had a higher value in predicting the survival and prognosis of PDAC.Conclusion:The five hub genes are significantly related to the prognosis of PDAC unearthed from the public biological database,which provides a theoretical basis for future research on the pathogenesis,clinical diagnosis and treatment targets of PDAC.
