MAP3K9在肺癌组织中的表达及其对肺癌细胞增殖、迁移、侵袭的影响

摘    要:
目的:检测MAP3K9在临床肺癌样本及肺癌细胞系中的表达,研究MAP3K9对肺癌细胞增殖、迁移、侵袭能力的影响,并结合受试者工作特征曲线(ROC曲线)评估其诊断价值。方法:从云南省肿瘤医院收集2018年09月至2019年12月经病理诊断确诊为肺癌患者的癌组织及其对应的癌旁组织。MAP3K9在肺癌组织及癌旁组织、肺癌细胞系A549和H1299、人正常肺上皮细胞系BEAS-2B中的mRNA表达量采用实时荧光定量PCR技术检测。MAP3K9在细胞系中的蛋白表达量采用蛋白质印迹技术检测。分别用CCK8、划痕和Transwell实验检测细胞的增殖、迁移、侵袭能力。利用在线生存数据库分析表达MAP3K9患者的生存率。使用SPSS软件绘制ROC曲线。结果:MAP3K9在肺癌组织中的表达量显著高于癌旁组织(P<0.05)。MAP3K9在A549和H1299中的表达量均高于BEAS-2B(P均<0.05)。体外细胞实验表明敲低MAP3K9能够明显抑制A549、H1299细胞的增殖、迁移和侵袭(P均<0.05)。生存曲线显示高表达MAP3K9的肺癌患者生存率低(P<0.05)。ROC曲线下面积为0.894(P<0.05),诊断临界值为1.36×10-3,诊断敏感度为71.2%,特异度为98.1%。结论:MAP3K9在临床肺癌样本及肺癌细胞系中高表达,该基因高表达能够促进肺癌细胞的增殖、迁移和侵袭,与患者预后不良有关,MAP3K9作为一个致癌基因有潜力成为临床诊断和治疗肺癌的有效靶点。

Abstract:
Objective:To detect the expression of MAP3K9 in clinical lung cancer samples and lung cancer cell lines,to investigate the effects of MAP3K9 on the proliferation,migration and invasion of lung cancer cells,and to evaluate its diagnostic value combining with receiver operating characteristic (ROC) curve.Methods:The cancerous tissues and corresponding paracancerous tissues of patients with lung cancer diagnosed by pathology from September 2018 to December 2019 were collected from Yunnan Cancer Hospital.The mRNA expression of MAP3K9 in lung cancer tissues and paracancerous tissues,lung cancer cell lines A549 and H1299,human normal lung epithelial cell line BEAS-2B was detected by real-time fluorescence quantitative PCR.The protein expression of MAP3K9 in cell lines was detected by Western blot.Cell proliferation,migration and invasion were evaluated by using cell counting kit 8 (CCK8),wound healing,Transwell assay,respectively.The survival rate of patients with MAP3K9 expression was analyzed by online survival database.ROC curve was drawn by SPSS software.Results:The expression of MAP3K9 in lung cancer tissues was significantly higher than that in paracancerous tissues (P<0.05).The expression of MAP3K9 in A549 and H1299 was higher than that of BEAS-2B (both P<0.05).In vitro cell experiments showed that knocking down MAP3K9 could significantly inhibit the proliferation,migration and invasion of A549 and H1299 cells (all of P<0.05).The survival curve showed that lung cancer patients with high MAP3K9 expression had lower survival rate (P<0.05).The area under ROC curve was 0.894 (P<0.05).The diagnostic threshold was 1.36×10-3.The diagnostic sensitivity was 71.2%,and the specificity was 98.1%.Conclusion:MAP3K9 is highly expressed in clinical lung cancer samples and lung cancer cell lines.The high expression of MAP3K9 can promote the proliferation,migration and invasion of lung cancer cells,which is related to the poor prognosis of patients.As an oncogene,it has the potential to become an effective target for clinical diagnosis and treatment of lung cancer.
