摘    要:目的:探究miR-194-5p调控LMNB1对肺腺癌细胞生长转移的作用。方法:实时荧光定量PCR(qRT-PCR)实验检测肺腺癌组织和癌旁组织中miR-194-5p、LMNB1表达水平。体外培养肺腺癌细胞A549,分为对照组、miR-194-5p mimics阴性对照组、miR-194-5p mimics组。转染处理后,CCK-8实验检测各组A549细胞增殖情况,比较各组细胞活力;流式细胞实验检测各组A549细胞凋亡率;细胞划痕及Transwell小室侵袭实验分别检测各组A549细胞迁移、侵袭力,比较各组细胞迁移、侵袭数;免疫印迹实验检测各组A549细胞凋亡蛋白caspase-3、Bax和上皮间质转化(epithelial-mesenchymal transition,EMT)相关蛋白E-cadherin、Vimentin表达;qRT-PCR实验及免疫印迹实验分别检测各组A549细胞miR-194-5p、LMNB1 mRNA表达及LMNB1蛋白表达。结果:相比癌旁组织,肺腺癌组织中miR-194-5p表达水平明显降低(P<0.05),LMNB1表达水平明显升高(P<0.05)。相比对照组,miR-194-5p mimics组A549细胞活力、细胞迁移数、细胞侵袭数、LMNB1 mRNA表达水平、Vimentin和LMNB1蛋白表达水平显著降低(P<0.05),细胞凋亡率、miR-194-5p表达、caspase-3、Bax和E-cadherin蛋白表达水平显著升高(P<0.05)。miR-194-5p mimics阴性对照组A549细胞上述各指标与对照组相比差异无统计学意义(P>0.05)。结论:miR-194-5p可下调LMNB1表达,抑制肺腺癌细胞增殖,促进其凋亡,并降低其迁移及侵袭能力。

Abstract:Objective:To explore the effect of miR-194-5p on the growth and metastasis of lung adenocarcinoma cells by regulating LMNB1.Methods:Real-time fluorescent quantitative PCR (qRT-PCR) was used to determine the expression level of miR-194-5p and LMNB1 in lung adenocarcinoma tissues and adjacent tissues.Lung adenocarcinoma cells A549 were cultured in vitro and divided into three groups:Control group,miR-194-5p mimics negative control group and miR-194-5p mimics group.After transfection,the CCK-8 experiment was used to detect the proliferation of A549 cells in each group,and compare the cell viability of each group.Flow cytometry assay was used to determine the apoptosis rate of A549 cells in each group.Cell scratch and Transwell invasion experiments were used to detect the migration and invasiveness of A549 cells in each group,and compare the numbers of cell migration and invasion in each group.Western blot experiment was performed to detect the expression of apoptotic proteins caspase-3,Bax and E-cadherin and Vimentin related to epithelial-mesenchymal transition (EMT) in each group of A549 cells.qRT-PCR experiment and Western blot experiment were used to detect the expression of miR-194-5p,LMNB1 mRNA and LMNB1 protein of A549 cells in each group.Results:Compared with adjacent tissues,the expression level of miR-194-5p in lung adenocarcinoma tissues was significantly reduced (P<0.05),and the expression level of LMNB1 in lung adenocarcinoma tissues was significantly increased (P<0.05).Compared with the control group,the A549 cell viability,cell migration number,cell invasion number,LMNB1 mRNA expression level,Vimentin and LMNB1 protein expression levels were significantly reduced in the miR-194-5p mimics group (P<0.05),and the apoptosis rate,miR-194-5p expression,caspase-3,Bax and E-cadherin protein expression levels increased significantly (P<0.05).There was no significant difference in the above indicators of A549 cells between the miR-194-5p mimics negative control group and control group (P>0.05).Conclusion:miR-194-5p can down-regulate the expression of LMNB1,inhibit the proliferation of lung adenocarcinoma cells,promote cells apoptosis,and reduce cells migration and invasion capabilities.
