Abstract:Objective To investigate the effect of electroacupuncture (EA) on the expression of brain-derived neurotrophic factor (BDNF), nerve growth factor (NGF), and acetylcholine esterase (AchE) in the retina and visual cortex of guinea pigs with lens-induced myopia (LIM).Methods Ninety two-week-old male guinea pigs were randomly divided into the normal control (NC) group, LIM group, and LIM+EA group. Guinea pigs in the NC group were bred normally without intervention, while guinea pigs in the LIM and LIM+EA groups were fitted with -6.0 D lenses on the right eye, and the left eye was used as self-control to establish a myopia model. Besides, guinea pigs in the LIM+EA group were given EA at the Temple and Hegu acupoints while wearing the lens. After 2 and 4 weeks of modeling, the diopter and axial length of guinea pigs in each group were measured, the mRNA and protein expression levels of BDNF, NGF, and AchE were determined by the quantitative polymerase chain reaction (qPCR) and enzyme-linked immunosorbent assay (ELISA), and the AchE vitality was detected by AchE assay kits.Results After 2 and 4 weeks of modeling, in terms of the difference between the model eye (right eye) and the self-control eye (left eye), the diopter and axial length in the LIM group were significantly increased compared with the NC group (both P<0.01), while the diopter and axial length in the LIM+EA group were significantly decreased compared with the LIM group (both P<0.05). qPCR and ELISA results showed that after 2 and 4 weeks of modeling, the mRNA and protein levels of BDNF and NGF in the retina and visual cortex of the right eye in the LIM group were significantly reduced compared with the NC group, while the mRNA and protein levels of AchE were significantly increased (all P<0.05). Compared with the LIM group, the mRNA and protein levels of BDNF and NGF in the retina and visual cortex of the right eye in the LIM+EA group were significantly increased, while the mRNA and protein levels of AchE were significantly decreased (all P<0.05). AchE assay results showed that after 2 and 4 weeks of modeling, the AchE vitality in the retina and visual cortex of the right eye in the LIM group was significantly higher than that in the NC group (all P<0.01). The AchE vitality in the retina and visual cortex of the right eye in the LIM+EA group was significantly lower than that in the LIM group (all P<0.01).Conclusion The BDNF and NGF levels in the retina and visual cortex of guinea pigs with myopia are decreased, while the AchE level is increased. EA can significantly elevate the expression levels of BDNF and NGF and decrease the expression level of AchE in myopic guinea pigs, thereby delaying the development of myopia.
