miR-127-3p通过靶向KIF3B抑制卵巢癌细胞增殖、侵袭及迁移

摘    要:目的:探讨miR-127-3p对卵巢癌细胞生物学行为的影响及其与KIF3B的关系。方法:选取SKOV3、HOSEpiC细胞。SKOV3细胞根据转染不同物质分为NC组、miR-NC组(miR-NC)、miR-127-3p组(miR-127-3p-mimics)、si-NC组(si-NC)和si-KIF3B组(si-KIF3B)。MTT评估增殖;Transwell分析迁移、侵袭;实时荧光定量PCR分析miR-127-3p、KIF3B mRNA含量;Western blot分析KIF3B蛋白表达。生物学、荧光素酶报告评估miR-127-3p与KIF3B的作用。结果:SKOV3细胞miR-127-3p水平低于HOSEpiC细胞(P<0.01),SKOV3细胞KIF3B蛋白、基因mRNA含量高于HOSEpiC细胞(P<0.01)。与NC组、miR-NC组相比,48和72 h时miR-127-3p组细胞增殖、迁移和侵袭能力下降(P<0.05),与NC组、si-NC组相比,48和72 h时si-KIF3B组细胞增殖、迁移和侵袭能力下降(P<0.05)。与NC、miR-NC组比较,miR-127-3p组KIF3B蛋白表达水平下降(P<0.01)。与WT-KIF3B组相比,miR-127-3p+WT-KIF3B组细胞荧光素酶活性下降(P<0.01)。结论:卵巢癌细胞低表达miR-127-3p,高表达KIF3B,miR-127-3p上调可能通过KIF3B抑制卵巢癌细胞的增殖、侵袭和迁移。

Abstract:Objective:To explore the effect of miR-127-3p on the biological behavior of ovarian cancer cells,and its relationship with kinesin family member 3B (KIF3B).Methods:SKOV3 and HOSEpiC cells were selected.SKOV3 cells were transfected and divided into NC group,miR-NC group (miR-NC),miR-127-3p group (miR-127-3p-mimics),si-NC group (si-NC) and si-KIF3B group (si-KIF3B).MTT was conducted to analyze proliferation ability.Transwell chamber test was conducted to analyze migration and invasion abilities.The expression of miR-127-3p and KIF3B mRNA in cells and tissues were detected by real-time quantitative PCR.Western blot was conducted to analyze the protein level of KIF3B.The relationship between miR-127-3p and KIF3B was predicted by bioinformatics and verified by luciferase reporter assay.Results:The level of miR-127-3p mRNA in SKOV3 cells were lower than that in HOSEpiC cells (P<0.01).The level of KIF3B protein and mRNA in SKOV3 cells were higher than that in HOSEpiC cells (P<0.01).Compared with NC and miR-NC groups,the proliferation ability at 48 and 72 h,the invasion and migration ability of miR-127-3p group were significantly decreased (P<0.05).Compared with NC and si-NC groups,the proliferation ability at 48 and 72 h,the invasion and migration ability of si-KIF3B group were significantly decreased (P<0.05).The protein level of KIF3B in miR-127-3p group was lower than that of NC and miR-NC groups (P<0.01).The luciferase activity of miR-127-3p+WT-KIF3B group was lower than that of WT-KIF3B group (P<0.01).Conclusion:Ovarian cancer cells have low expression of miR-127-3p and high expression of KIF3B.Overexpression of miR-127-3p may inhibit the proliferation,invasion and migration through KIF3B.
