摘    要:目的:探讨长链非编码RNA(long non-coding RNA,lncRNA)LOC572558对恶性黑素瘤细胞增殖、迁移及侵袭能力的影响。方法:使用定量实时聚合酶链式反应(qRT-PCR)检测LOC572558在20对恶性黑素瘤及癌旁组织中以及在NHEM、A375、A2058和B16细胞系中的表达。应用小干扰 RNA(siRNA)技术下调 LOC572558表达后,在A375和B16细胞系中使用流式细胞术评估细胞凋亡情况,划痕实验检测细胞的迁移能力,Transwell小室法检测细胞的侵袭能力。结果:lncRNA LOC572558在恶性黑素瘤组织及细胞系中高表达,且下调lncRNA LOC572558表达可以促进A375和B16细胞的凋亡,抑制A375和B16细胞的迁移及侵袭能力。结论:lncRNA LOC572558有望成为临床早期诊断恶性黑素瘤潜在分子标志物以及治疗晚期恶性黑素瘤的靶向标志物。

Abstract:Objective:To investigate the effects of long non-coding RNA (lncRNA) LOC572558 on the proliferation,migration and invasion of malignant melanoma cells.Methods:Quantitative real-time polymerase chain reaction (qRT-PCR) was used to detect the expression of LOC572558 in 20 pairs of malignant melanoma and its paired adjacent tissues,as well as in NHEM,A375,A2058 and B16 cell lines.The expression of LOC572558 in A375 and B16 cells was knocked down by small interfering RNA (siRNA),and flow cytometry was used to assess cell apoptosis in cell lines.The cell migration ability was detected by scratch test,and Transwell cell assay was used to study the invasion ability of cells.Results:lncRNA LOC572558 was highly expressed in melanoma tissues and cell lines,and down-regulation of lncRNA LOC572558 expression can significantly promote apoptosis,inhibit migration and invasion of A375 and B16 cell lines.Conclusion:lncRNA LOC572558 is expected to be a potential molecular marker for early clinical diagnosis of malignant melanoma and a targeted marker for the treatment of advanced malignant melanoma.
