抑癌基因p16对人肺腺癌裸鼠移植瘤的生长抑制

摘    要:目的 观察不同途径给予抑癌基因 p16对人肺腺癌裸鼠移植瘤的治疗效果。 方法 分别将转染和未转染p16基因的肺腺癌细胞系H4 6 0接种于裸鼠背部皮下 (p16治疗B组和模型组 ) ,再将模型组裸鼠 18只随机分为空白对照组 ,脂质体对照组及p16基因治疗A组。瘤内分别注射生理盐水、脂质体以及脂质体 p16混合物 ,定期测量瘤体体积 ,连续观察 6周 ,免疫组化检测荷瘤组织 p16蛋白的表达。结果 p16基因治疗A、B组其瘤体生长速度明显减缓 ,与对照组相比 ,差异有显著性意义 (P <0 0 1,P <0 0 5 ) ,p16基因治疗B组生长受抑作用更为显著。免疫组化结果提示两治疗组 p16蛋白表达阳性率高于两对照组。结论 脂质体介导的 p16基因转染可抑制人肺腺癌裸鼠移植瘤的生长。但与瘤体内直接转染比较 ,先转染再致瘤的抑制作用更强。

Abstract:Objective To investigate the effect of tumor suppressor gene p16 on the nude mice models of lung adenocarinoma by different introduction ways. Methods Nude mice were injected with human lung adenocarcinoma cell line H460 or H460 cell transfected with p16 gene respectively (p16 treatment B group and model groups). After 2 weeks, 18 nude mice implanted with tumors in model group were randomly divided into 3 groups (control, lipofectin, p16 treatment A groups) and subjected to the injection of normal saline, lipofectin and the mixture of lipofectin and p16 respectively. The size of tumor was measured after treatment. The expression of p16 protein in implanted tumor was detected by using immunohistochemistry. Results The growth of tumor in both p16 treatment groups (group A and B) were obviously slower down as compared with the control groups (P<0.01 or P<0.05). The inhibitive effects on growth were more remarkable in group B. The expression of p16 protein in both p16 treatment groups was significantly higher than that in the controls. Conclusion The transfected p16 gene induced by lipofectin can inhibit the growth of implanted tumor on nude mice. Induction of p16 gene in vitro have a greater antitumor effect.
