卵巢上皮性肿瘤组织中PI3K和p-AKT蛋白的表达

摘    要:目的:探讨3羧基磷脂酰肌醇激酶(PI3K)和磷酸化蛋白激酶B(p-AKT)在上皮性卵巢肿瘤组织中的表达、相互关系及其意义.方法:应用免疫组化方法,检测20例正常卵巢、20例卵巢良性上皮肿瘤、60例卵巢癌组织中PI3K和P-AKT蛋白的表达.结果:PI3K在卵巢癌组织中阳性表达率为58.3%,高于正常卵巢和良性上皮肿瘤组织的5%,20%(P〈0.01).p-AKT在卵巢癌组织中阳性表达率为53.3%,高于正常卵巢和良性上皮肿瘤组织的10%,20%(P〈0.01).PI3K和p-AKT蛋白表达与卵巢癌临床分期、病理分级、有无淋巴结转移和远处转移有关(P均〈0.05),而与年龄、病理类型、有无腹水无关(P均〉0.05).卵巢癌组织中PI3K和p-AKT蛋白间的表达呈正相关(rs=0.501,P〈0.001).结论:AKT是PI3K的一个重要靶基因,PI3K/AKT信号传导通路异常活化与卵巢癌的发生、发展有关.

Abstract:AIM: To investigate the expressions of phosphatidylinositol 3-kinase(PI3K) and phosphorylated-AKT(p-AKT) proteins in epithelial ovarian cancers and their correlations. METHODS: Expressions of PI3K and p-AKT in 20 normal ovarian tissues, 20 benign tumors and 60 ovarian epithelial cancers were detected by immunohistochemical method. RESULTS: The positive rates of PI3K in normal ovarian tissues (5%)and benign tumor tissues (20%)were significantly lower than that in epithelial ovarian cancers(58.3%, P<0.01). The positive rates of p-AKT in normal ovarian tissues(10%) and benign tumor tissues (20%)were significantly lower than that in ovarian epithelial cancers(53.3%, P<0.01). The expressions of PI3K and p-AKT proteins were correlated to clinical stages, differentiation degree, lymph node metastasis and distant metastasis(P<0.05). but not to age,histological type and ascites(P>0.05). In ovarian cancers, a positive correlation was observed between the expressions of PI3K and p-AKT (rs=0.501, P<0.001). CONCLUSION: AKT is an important target gene of PI3K.The abnormality of PI3K/AKT pathway is associated with the development of epithelial ovarian cancer.
