eIF3b在三阴性乳腺癌细胞增殖和侵袭中的作用

摘    要:目的:探讨真核细胞翻译起始因子3的亚基b(eIF3b)基因在三阴性乳腺癌(TNBC)增殖和侵袭中的作用。方法:采用免疫组化法检测50例TNBC组织中eIF3b蛋白的表达;应用实时定量聚合酶链反应(RT-q PCR)、细胞计数试剂盒8(CCK-8)、细胞克隆形成、细胞划痕实验、细胞侵袭实验,以及蛋白质印迹(Western blot)法分别检测RNA干扰(RNAi)前后MDA-MB-231细胞株eIF3b mRNA表达、细胞增殖、细胞迁移、细胞侵袭能力的变化,以及β-Catenin、C-myc蛋白的表达。结果:50例TNBC中eIF3b高表达率为74%,显著高于癌旁正常组织(P<0.01),与Ki-67表达、淋巴结转移有关(P<0.05),而与患者年龄、肿瘤大小、组织学类型无关(P>0.05)。RNAi后MDA-MB-231细胞株的eIF3b mRNA水平显著下降(P<0.05),细胞的增殖、迁移和侵袭能力显著低于对照组(P<0.05),Western blot结果显示β-Catenin蛋白和C-myc蛋白表达水平显著低于对照组(P<0.05)。结论:e...

Abstract:Objective: To explore the effect of eukaryotic translation initiation factor 3 subunit b (eIF3b) on proliferation and invasion of triple-negative breast cancer (TNBC). Methods: Immunohistochemistry was used to detect the expression of eIF3b in 50 cases of TNBC. RNA interference (RNAi) was accomplished by transfecting eIF3b-siRNA into the cell line MDA-MB-231. The mRNA expression level of eIF3b was detected using RTqPCR.The proliferation and invasion of cells was detected using the cells proliferation rate detected by CCK-8, cell clone formation, the cell scratch test and Transwell invasion assay. The expression of β-Catenin and Cmyc was detected by Western blot. Results: The high expression rate of eIF3b in 50 cases of TNBC was 74%,significantly higher than adjacent normal tissues (P<0.01). eIF3b expression was significantly related to Ki-67 expression and lymphatic metastasis (P<0.05), but not related to age, tumor size, type of histopathology (P>0.05).The mRNA expression of eIF3b and the potential for cell proliferation, migration and invasion were significantly lower in siRNA transfected cells than that in non-transfected cells (P<0.05). The results of Western blot showed significantly decreased (P<0.05) expression of β-Catenin and C-myc. Conclusion: eIF3b is highly expressed in TNBC. Downregulation of eIF3b expression could inhibit cell proliferation, migration and invasion, which may be attributed to disordered cell cycles by blocking Wnt/β-Catenin signal pathway.
