子宫内膜异位症相关巨噬细胞的提取和鉴别

摘    要:目的:探究分离提取子宫内膜异位症(EMs)相关巨噬细胞的方法。方法:免疫组织化学法检测正常子宫内膜切片以及EMs切片中CD68和CD163的表达。收集3例经腹腔镜确诊为EMs患者病灶,通过机械法结合IV型胶原酶消化法分离EMs相关巨噬细胞,并利用流式细胞术检测细胞CD11b、CD163、CD80的表达情况。结果:EMs组织中可见2型巨噬细胞高表达,提取的EMs原代巨噬细胞具备巨噬细胞的形态特征,其中CD11b和CD163双阳性比例为6.46%,且未见CD80阳性细胞表达。结论:本研究采用的方法可有效提取EMs组织相关巨噬细胞,提高了纯度,对进一步研究EMs相关巨噬细胞特性及其生物学行为具有重要价值。

Abstract:Objective: To explore the method of extracting endometriosis (EMs)-associated macrophages (EAMs). Methods: Immunohistochemistry staining was used to explore the expression of CD68 and CD163 in pathological sections of ectopic endometrium (Ec) and normal endometrium (En). Collect three cases of EMs patients diagnosed by laparoscopy, separate EAMs by mechanical method combined with type IV collagenase digestion, and use flow cytometry to detect the expression of CD11b, CD163, and CD80. Results: High expression of type 2 macrophages can be seen in EMs tissues. The extracted EMs primary macrophages have the morphological characteristics of macrophages. The double-positive ratio of CD11b and CD163 is 6.46%, and no CD80-positive cell expression is seen. Conclusion: The method used in this study can effectively extract EAMs and improve their purity, which is of great value for further research on the characteristics and biological behavior of endometriosis-associated macrophages.
