2型糖尿病并脂肪肝实验性大鼠模型的建立及评价

[目的]建立发病机制与人类相似的2型糖尿病并脂肪肝的动物模型.[方法]Wistar大鼠随机分为正常组、链脲佐菌素(STZ)25 mg/kg加高脂饲料组(先注射STZ后喂以高脂饲料)、STZ 12.5 mg/kg加高脂饲料组、高脂饲料加STZ 25 mg/kg组(先喂以高脂饲料后注射STZ)、高脂饲料加STZ 12.5 mg/kg组.10周后,测动物血糖、血脂、肝功能、胰岛素;取肝切片,苏木精-伊红染色、苏丹三醇染色.[结果]STZ 25 mg/kg加高脂饲料组动物血糖、血脂、肝功能、胰岛素抵抗指数有显著的变化,组织病理学也有典型脂肪肝变化.[结论]小剂量STZ注射加高脂饲料喂养可以建立2型糖尿病并脂肪肝动物模型.

Establishing and valuating animal model resembling to type two diabetes mellitus with intercurrent fatty liver

[Objective] To establish the animal model resembling to type two diabetes mellitus with intercurrent fatty liver.[Methods] Wistar rats were divided into five groups: normal group(group A),25mg/kg STZ adding high-fat-feed group(group B,feeding with high fat after injecting STZ),12.5 mg/kg STZ adding high-fat-feed group(group C),high-fat-feed adding 25 mg/kg STZ group(group D,injecting STZ after feeding with high fat) and high-fat-feed adding(12.5 mg/kg) STZ group(group E).Ten weeks latter,blood glucose(BG),triglyceride(TG),cholesterol(CHO),high density lipoprotein(HDL) and insulin(RI) were examined and analyzed,and liver tissue was sliced up and stained with hematoxylin and eosin or sudantriol.[Results] There were markedly changes in BG,TG,CHO,HDL,insulin resistance index and similar histopathology to fatty liver in group B.[Conclusion]The way of lower STZ injection adding high fatfeed can establish the animal model of type two diabetes mellitus with intercurrent fatty liver.
