摘    要:目的:探讨抑癌基因PTEN蛋白在人乳腺癌中的表达情况,及其与血管内皮生长因子(VEGF)表达的相关性。方法:应用免疫组织化学染色S-P法,检测44例乳腺癌组织石蜡切片中PTEN蛋白、VEGF蛋白的表达情况,比较PTEN蛋白表达与临床病理指标的关系,及其与VEGF蛋白表达的相关性。结果:PTEN蛋白表达定位于细胞质。PTEN蛋白表达减低者占47.7%(21/44),阳性表达者占52.3%(23/44)。PTEN蛋白表达与乳腺癌是否淋巴结转移、乳腺癌雌激素状态及局部是否复发相关(P<0.05)。PTEN与VEGF蛋白表达呈负相关,但无统计学意义(r=-0.236,P=0.122)。结论:乳腺癌患者PTEN蛋白低表达与预后不良有关,PTEN对乳腺癌有一定的预后价值。在人乳腺癌中,VEGF可能是PTEN的主要下游基因。

Abstract:Objective To observe the expression and significance of PTEN and VEGF in breast cancer and benign breast disease, and their relation with the tumor size, histological grade, the level of estrogen receptor, axillary lymphatic metastasis and relapse. Methods PTEN and VEGF protein expressions were detected by immunohistochemical S- P method in paraffin sections from women with primary breast cancer. PTEN protein expression was compared with clinical pathologic parameters and VEGF protein expression. Results In the breast cancer, PTEN, being shown in the cytoplasm, was reduced in 47.7% (21/44) and positive in 52.3% (23/44) samples, while all 10 cases of benign breast disease showed positive staining of PTEN(P< 0.01). The level of PTEN expression was correlated with axillary lymph node status(P< 0.01), loss of estrogen receptor stain(P< 0.05), and recurrence(P< 0.05). Patients with positive PTEN were negatively with VEGF in breast cancer(r=- 0.236,P=0.122).Conclusion PTEN, whose lower expression is correlated with poor outcome of breast cancer, may play a prominent role in breast cancer. VEGF may be the primary downstream genes of PTEN in breast cancer.
