| 摘 要： | 目的构建过表达iASPP的乳腺癌细胞株，分析过表达iASPP对顺铂（cis-dichlorodiamine platinum，DDP）作用下乳腺癌细胞的影响。方法采用RT-PCR方法在乳腺组织中扩增出iASPP，转染人MCF-7，G418筛选获得过表达iASPP的乳腺癌细胞株，MTT和流式细胞仪分析过表达iASPP的乳腺癌细胞株在DDP作用下增殖和凋亡情况。结果成功构建了过表达iASPP的乳腺癌细胞株，过表达iASPP可明显降低DDP对乳腺癌细胞增殖的抑制作用（P〈0．01），并降低细胞凋亡的百分数（P〈0．01）。结论过表达iASPP可以明显降低乳腺癌细胞MCF-7对DDP作用的敏感性。

| Abstract: | Objective To analyze the effects of overexpressed iASPP on breast cancer cell line MCF-7. Methods The iASPP cDNA was obtained from breast carcinoma tissue by RT- PCR. The breast cancer cell line MCF-7 that overexpressed iASPP was established by stable transfection and G418 selection. The proliferation and apoptosis of the MCF-7 cells that overexpressed iASPP after exposure to cisplatin were detected by MTT and flow cytometry.
