硫酸皮肤素对大鼠血管纤溶酶原活化素释放的影响

摘    要:用大鼠下肢血管床灌流模型,观察肝素样药物对纤维蛋白溶解系统的作用。结果表明,硫酸皮肤素(DS)能诱导大鼠肢体血管壁释放纤溶酶原活化素(PA)。DS浓度0.1~0.8mg/ml时,给药后1~2min内PA活性增高达最大值。DS在0.1~0.4mg/ml范围内时PA释放与DS浓度成正比,DS0.4mg/ml可诱导最大的PA释放反应。SDS-聚丙烯酰胺凝胶电泳和纤维蛋白自显影分析证实,DS诱导的是组织类型纤溶酶原活化素(t-PA),其分子量为67000。

Abstract:Effect of dermatan sulphate (DS) on plasminogen activator (PA) activity was studied in rat perfusion model. Perfusion of different doses of DS (0.1-0.8 mg/ml) resulted a increase of PA activity which reached the maximum within the 1-2 min after perfusion, gradual increase of PA activity being noticed within a certain concentration range of DS (0.1-0.4 mg/ml) and decrease thereafter (0.6-0.8mg/ml). The type of PA activity increase during DS perfusion was characterized by SDS-PAGE and fibrinautography as tissue -type PA (t-PA) on the basis of its molecular weight (67 000) which could be inhibited by a specific anti-t-PA antiserum.
